苯酚-硫酸法测定百蕊草中多糖的含量

摘　　要：目的 建立苯酚-硫酸法测定百蕊草多糖的含量。方法 采用60％乙醇除去黄酮类成分，再以水提醇沉的方法分离纯化百蕊草多糖，最后用苯酚-硫酸法测定百蕊草多糖的含量。结果 在0．01—0.07mg／mL内，浓度与吸收度呈良好线性。回归方程：A=15．644C-0．0053，r=0．9995，平均回收率为99．6％，RSD为1．92％（n=9）。百蕊草多糖的含量为15．9％，RSD为1．56％（n=5）。结论 方法简便，结果准确，为百蕊草多糖的进一步研究奠定良好的基础。OBJECTIVE To develop phenol-sulfuric acid method for the determination of polysaccharide in Thesium chinense Turcz. METHODS The flavones in Thesium chinense turcz, were eliminated with 60% ethanol, then the polysaccharide of Thesium chinense Turcz. was separated and purified with water extracting and ethanol depositing. The content was determined by phenol-sulfuric acid method. RESULTS From the range of 0.01 - 0.07mg/mL,tbere was an excellent linear shape between the concentration and absorptance. The regression equation ：A = 15. 644C - 0. 0053 ,r = 0.999 5. The average recovery was 99.6% with RSD of 1.92% （ n = 9）. The content was15.9%, RSD = 1.56% （ n = 5 ）. CONCLUSION The method is simple and accurate.
