摘  要:目的 建立苯酚-硫酸法测定百蕊草多糖的含量。方法 采用60%乙醇除去黄酮类成分,再以水提醇沉的方法分离纯化百蕊草多糖,最后用苯酚-硫酸法测定百蕊草多糖的含量。结果 在0.01—0.07mg/mL内,浓度与吸收度呈良好线性。回归方程:A=15.644C-0.0053,r=0.9995,平均回收率为99.6%,RSD为1.92%(n=9)。百蕊草多糖的含量为15.9%,RSD为1.56%(n=5)。结论 方法简便,结果准确,为百蕊草多糖的进一步研究奠定良好的基础。OBJECTIVE To develop phenol-sulfuric acid method for the determination of polysaccharide in Thesium chinense Turcz. METHODS The flavones in Thesium chinense turcz, were eliminated with 60% ethanol, then the polysaccharide of Thesium chinense Turcz. was separated and purified with water extracting and ethanol depositing. The content was determined by phenol-sulfuric acid method. RESULTS From the range of 0.01 - 0.07mg/mL,tbere was an excellent linear shape between the concentration and absorptance. The regression equation :A = 15. 644C - 0. 0053 ,r = 0.999 5. The average recovery was 99.6% with RSD of 1.92% ( n = 9). The content was15.9%, RSD = 1.56% ( n = 5 ). CONCLUSION The method is simple and accurate.
