摘 要:目的 建立高效液相色谱法测定珍菊降压片中盐酸可乐定、氢氯噻嗪的含量及含量均匀度。方法 采用C18柱(2.50mm×4.6mm,5μm),测定盐酸可乐定的流动相为乙腈-0.2%磷酸(5:95)、检测波长为210nm;测定氢氯噻嗪的流动相为甲醇-0.1%磷酸(15:85),检测波长为332nm;流速1mL/min。结果 线性范围:盐酸可乐定7.132~142.64ng(r^2=1.0000);绿原酸0.0511~0.61321μg(r=0.9999);氢氯噻嗪1.0023~12.02761μg(r=1.0000)。加样回收率:盐酸可乐定为99.5%,RSD为2.7%(n=9);绿原酸为99.7%,RSD为1.57%(n=9);氢氯噻嗪为100.4%,RSD为1.03%(n=9)。结论 本方法操作简单、结果准确,可作为控制本品质量的方法。OBJECTIVE To develop HPLC methods for determination of clonidine hydrochlofide, hydrochlorothiazide and chlorogenie acid uniformity in Zhenju Jiangya tablets. METHODS The HPLC analysis was performed on a Agilent Zorbax Extend C18 (250mm×4.6mm ,5μm), elonidine hydrochloride was separated using the mobile phase consisted of acetanilide and 0.2% H3 PO4 (5: 95), detected at 210nm, hydrochlorothiazide and ehlorogenie acid were done using the mobile phase consisted of methanol and 0.1% H3PO4 (15:85 ), monitored at 332nm, flow rate was 1.0mL·min^-1. RESULTS The linearity ranges of elonidine hydrochloride, chlorogenic acid and hydrochlorothiazide were 7. 132 - 142.64ng (r^2 = 1. 000 0), 0. 0511 -0. 61320g (r = 0. 999 9 ), 1. 0023 - 12.0276μg( r = 1. 000 0), the recovery rate of analytical method was 99.5% ,RSD was 2.2% ( n =5) for elonidine hydrochloride, was 99.7% , RSD was 0.92% ( n = 6) for chlorogenic acid, was 100.4% , RSD was 1.03% ( n = 6) for hydrochlorothiazide, respectively. CONCLUSION The methods developed are simple and accurate, and can be used to control the quality of Zhenju Jiangya tablets.
