摘  要:目的 对比研究发酵虫草菌粉中甘露醇的提取和后处理方法,建立甘露醇含量的比色测定法。方法 采用超声提取、高速离心分离进行样品预处理,比色法测定发酵虫草菌粉中甘露醇的含量。结果 甘露醇的线性范围为20~90μg/mL,加样回收率为102.6%,RSD为1.34%。重复性实验的RSD为0.44%,供试品溶液在24h内稳定。2001年生产的发酵虫草菌粉中甘露醇含量的平均值比前2年约高2%,含量变化范围比前两年稍小,3年内生产的各批次甘露醇含量波动均在10%左右,发酵虫草菌粉质量较为稳定。结论 用超声离心比色法测定甘露醇含量,简捷快速,重现性好,回收率高,可以用于发酵虫草菌粉的质量控制。OBJECTIVE To select the optimal method of extracting mannitol from Cordyceps sinensis ( Berk. ) Sacc. for determining the content of mannitol in the herb using colourimetric method. METHODS Ultrasonic extraction following centrifugation was used for extracting the active compound from the herb. RESULTS The linear range of mannitol was 20 - 90μg/mL and the recovery was 102.6% with RSD 1.34% (n =5 ). Variation coefficient was 0.44% and the sample solution was stable in 24 h. The proposed method was applied to determine the content of mannitol in 60 batches of fermented Cordyceps sinensis ( Berk. ) Sacc. produced in the years of 1999, 2000 and 2001. The average concentration of mannitol in the fermented herb produced in 2001 was about 2% higher than those in 1999, 2000 and with less remarkable variation, the content of mannitol in 60 batches of the herb produced in the 3 years was found to fluctuate at 10%. CONCLUSION The proposed method is effective and convenient for the analysis of mannitol in quality control of Cordyceps sinensis ( Berk. ) Sacc..
