摘  要:目的探讨猫脊髓半横断损伤后急性期移植小鼠胚胎干细胞后存活、迁移及对功能的影响.方法胚胎干细胞移植前进行培养诱导和记数绿色荧光蛋白标记的胚胎干细胞.取成年健康猫6只(正常移植组和半横断移植组,每组各3只),其中半横断组实施脊髓半横断手术.正常组和半横断组同时移植带有绿色荧光小鼠胚胎干细胞,7 d后灌注取材并行脊髓组织切片后,在倒置荧光显微镜下观察干细胞存活及迁移情况和对行为功能的影响.结果绿色荧光蛋白标记的胚胎干细胞的阳性率为38%,半横断移植组未见到荧光细胞,正常移植组可看到荧光细胞并迁移一段距离.结论小鼠胚胎干细胞能在成年猫的脊髓存活、迁移.但半横断脊髓损伤组未见荧光细胞,提示炎症急性期不利于移植的干细胞存活或者免疫排斥反应导致干细胞死亡.Objective To investigate the survival, migration and function of mouse embryonic stem cells (ESCs) implanted into spinal cord of cats at acute stage following spinal cord hemisection injury. Methods ESCs were collected, induced and marked with green fluorescent protein (GFP) before transplantation. 6 healthy adult cats were randomly divided into 2 groups, normal transplanted group and spinal cord hemisection transplanted group ( n = 3/each group) . After spinal cord hemisection injury, GFP- ESCs were transplanted immediately into spinal cord. All rats were perfused 7 days later. The sections were observed under fluorescence microscope. Results The positive rate of ESCs with GFP is 38%. Fluorescence ceils and the migration distance were seen in normal transplanted group, not in hemisection transplanted group. Conclusion The acute inflammation stage might not facilitate the survival of the implanted cells or immunological rejection resuhed in the death of ESCs, indicating it should be considered for the transplantation time of ESCs after inflammation stage.
