摘  要:以酶学分析方法研究了10种金属离子(Ag+、Cd2+、Ca2+、Cu2+、Fe3+、Hg2+、Mg2+、Mn2+、Pb2+和Zn2+)对三疣梭子蟹Portunus trituberculatus中肠腺蛋白酶、胰蛋白酶、胰凝乳蛋白酶、羧肽酶A、羧肽酶B、淀粉酶和纤维素酶活性的影响,10种金属离子浓度分别设置为5×10-4、1×10-3和5×10-3mol.L?13个梯度。结果表明,在实验浓度范围内低浓度(0.5mmol.L-1)Fe3+、Pb2+、Mn2+及0.5和1mmol.L-1的Ca2+对蛋白酶活性影响不显著,Mg2+对蛋白酶的激活作用随浓度的升高而升高,1mmol.L-1的Mn2+对蛋白酶表现为显著激活作用;3种浓度的Ca2+、Cd2+、Mg2+、Mn2+对胰蛋白酶均表现显著激活作用;Ca2+及低浓度的Mg2+和Cu2+对胰凝乳蛋白酶有激活作用;Hg2+及低浓度的Mg2+、Mn2+、Zn2+对羧肽酶A有激活作用;Ca2+及低浓度的Mg2+对羧肽酶B有激活作用;Ca2+、Cd2+、Mn2+和低浓度的Zn2+、Fe3+、Pb2+对淀粉酶表现为激活作用;Zn2+、Fe3+、Mn2+、Cu2+对纤维素酶表现为激活作用。除此之外,金属离子对这几种消化酶均表现为抑制作用,抑制作用随浓度的升高而升高。Portunus trituberculatus was bought from a market in Qingdao, with body weigh of 99. 75±5.2 g, and fed in plastic tanks for 7 days. The midgut gland was removed after 48h starvation, then the effects of ten metal ions (Ag^+ , Cd^2+ , Ca^2+ , Cu^2+ , Fe^3+ , Hg^2+ , Mg^2+ , Mn^2+ , Pb^2+ and Zn^2+ ) at different con- centrations (5×10^-4 , 1 × 10^-3 , and 5 × 10^-3 mol · L^-1 respectively) on the activities of protease, trypsin, chymotrypsin, carboxypeptidase A, carboxypeptidase B, amylase, and cellulase of the midgut gland of Portunus trituberculatus were studied by using enzyme analytical method. The experimental results indicated that Fe^3+ , Pb^2+ and Mn^2+ at 0.5 mmol·L^-1 and Ca^2+ at 0.5 mmol·L^-1 and lmmol·L^-1 had no significant effects on protease; Mn^2+ at 1 mmol·L^-1 had significant activation on protease, and the activation of Mg^2+ on protease activity increased with its concentration increasing; Ca^2+ , Cd^2+ , Mg^2+ and Mn^2+ at the three concentrations all had significant activation on trypsin; Chymotrypsin was activated by Ca^2+ and low concentrations of Mg^2+ and Cu^2+ ; Carboxypeptidase A activity was enhanced by Hg^2+ and low concentrations of Mg^2+ , Mn^2+ and Zn^2+ ; Carboxypeptidase B activity was enhanced by Caz+ and low concentrations of Mg^2+ ; Amylase activity was enhanced by Ca^2+ , Cd^2+ , Mn^2+ and low concentrations of Zn^2+ , Fe^3+ and Pb^2+ ; Cellulase activity was enhanced by Zn^2+ , Fe^3+ , Mn^2+ and Cu^2+. In addition, these digestive en-zymes were all inhibited by the metal ions, and the inhibition was intensified with concentration increasing.
