摘  要:采用1%羧甲基茯苓多糖(CMP)腹腔注射,摘取乙型肝炎病毒(HBV)转基因小鼠脾脏,制备淋巴细胞,MTT(噻唑蓝粉剂)法观察CMP对淋巴细胞的毒性作用;经粒-巨噬细胞集落刺激因子(GM-CSF)和白细胞介素-4(IL-4)培养转基因小鼠脾脏来源树突状细胞(DC),ELISA法检测DC分泌IL-12以及刺激混合淋巴细胞反应T淋巴细胞分泌IL-10、IFN-γ的含量.CMP在0~500mg/L浓度对HBV转基因小鼠无毒性作用,并能显著促进HBV转基因小鼠DC分泌IL-12,在混合淋巴细胞反应中,能显著促进T淋巴细胞分泌IFN—γ并抑制IL-10的分泌,从而上调DC功能.Carboxymethytl pachymaram of 1% concentration was given to HBV transgenic mice by abdominal injection. Lymphocytes were made from spleens. MTT method was used to detect CMP's cytotoxicity. Dendritic cells were separated from lymphocytes and incubated with granulocyte macrophage colony-stimulating factor (GM-CSF)and interleukin-4(IL-4). Phenotype of DCs were assayed by flowcytome try(FCM) ,IL-12 released by DCs and IL-10 and IFN-γ produced by T cells in mixed lymphocyte reaction (MLR) were measured by ELISA. Results showed that CMP within the concentrations of 0-500 μg/ml didn't show cytotoxicity to lymphocytes, CMP could enhance DCs' phenotype, increase IL-12 level in HBV transgenic mice ,increase the secretion of IFN-γ,inhibit the secretion of IL-10 in MLR,thus up-regulates DC's function.
