补肾健脾养血活血方对老年小鼠肾线粒体DNA缺失突变的影响

摘　　要：目的:研究补肾健脾养血活血方对老年Balb/c小鼠肾线粒体DNA缺失突变的影响。方法:老年Balb/c小鼠随机分为老年空白组和老年给药组,分别给予生理盐水和补肾健脾养血活血方4个月。采用聚合酶链反应(PCR)技术和光密度扫描检测两组线粒体DNA的缺失突变情况。结果;与老年空白组比较,补肾健脾养血活血方能显著减少老年Balb/c小鼠肾mtDNA的缺失(P<0.001)。结论:补肾健脾养血活血方可以抑制老年小鼠mtDNA的缺失突变,提示补肾健脾养血活血法组方能减少mtDNA的氧化损伤,对mtDNA有保护作用,从而从分子水平提供了本方延缓衰老的可能机制。Objective： To study the effect of Bushen Jianpi Yangxue Huoxue Formula （BJYHF） on deletion mutation of kidney mitochondria DNA in senile mouse. Methods： Senile Balb/c mice were randomly divided into senile blank group （SBG） and senile medication administration group （SMAG）, given normal saline and BJYHF for 4 months respectively. Deletion mutation state of the two groups of mitochondria DNA were detected by PCR technology and photodeusity scan. Results： Compared with SBG, BJYHF could decrease depletion of kidney mtDNA in senile Balb/c mouse significantly （ P 〈 0.001）. Conclusion： BJYHF can inhibit deletion mutation of mtDNA in senile Balb/c mouse, h suggests that BJYHF can reduce oxidative damage of mtDNA and protect mtDNA. Accordingly it provides the possible mechanism of anti - aging effect of BJYHF from the molecular level.
