摘  要:目的:研究补肾健脾养血活血方对老年Balb/c小鼠肾线粒体DNA缺失突变的影响。方法:老年Balb/c小鼠随机分为老年空白组和老年给药组,分别给予生理盐水和补肾健脾养血活血方4个月。采用聚合酶链反应(PCR)技术和光密度扫描检测两组线粒体DNA的缺失突变情况。结果;与老年空白组比较,补肾健脾养血活血方能显著减少老年Balb/c小鼠肾mtDNA的缺失(P<0.001)。结论:补肾健脾养血活血方可以抑制老年小鼠mtDNA的缺失突变,提示补肾健脾养血活血法组方能减少mtDNA的氧化损伤,对mtDNA有保护作用,从而从分子水平提供了本方延缓衰老的可能机制。Objective: To study the effect of Bushen Jianpi Yangxue Huoxue Formula (BJYHF) on deletion mutation of kidney mitochondria DNA in senile mouse. Methods: Senile Balb/c mice were randomly divided into senile blank group (SBG) and senile medication administration group (SMAG), given normal saline and BJYHF for 4 months respectively. Deletion mutation state of the two groups of mitochondria DNA were detected by PCR technology and photodeusity scan. Results: Compared with SBG, BJYHF could decrease depletion of kidney mtDNA in senile Balb/c mouse significantly ( P 〈 0.001). Conclusion: BJYHF can inhibit deletion mutation of mtDNA in senile Balb/c mouse, h suggests that BJYHF can reduce oxidative damage of mtDNA and protect mtDNA. Accordingly it provides the possible mechanism of anti - aging effect of BJYHF from the molecular level.
