雷公藤内酯醇、顺铂对骨肉瘤细胞体外增殖的影响

摘　　要：目的 研究中药制剂雷公藤内酯醇（TP）和顺铂（PDD）对骨肉瘤（MG63）细胞增殖的抑制作用，寻求骨肉瘤的中西医结合治疗方案。方法 将TP与PDD单独及联合于MG63细胞．采用MTT法检测细胞抑制率。结果 TP在1～125μg／ml的范围内对MG63细胞生长有明显抑制作用；PDD在1～20μg／ml的范围内对MG63细胞生长有明显的抑制作用；TP1～15μg／ml和PDD1μg／ml联合对MG63细胞生长有明显抑制作用；与对照组比较。P均〈0．01；TP10～15μg／ml和PDD1μg／ml联合细胞抑制率明显高于单用；二者单独及联合对细胞抑制率的影响均有明显时间依赖性。结论TP与PDD单独及联合均可抑制骨肉瘤细胞的增殖。[Objective ] To investigate the proliferation inhibitory effect of triptolide and cisplatin on osteosarcoma cells, so as to explore the new method of clinical therapy, [Methods] Triptolide and cisplatin were applied on the osteosarcoma cells MG63 respectively or jointly and the inhibition rate was measured by MTT assay. [Results] 1-125 μg/ml triptolide significantly inhibited the proliferation of MG63 cells （P〈0. 01）; 1-20μg/ml cisplatin significantly inhibited proliferation of MG63 cells （P 〈 0. 01） ; 1 -15 μ/ml triptolide combined with 1 μg/ml cisplatin had a marked inhibitory effect on MG63 cells （P〈0. 01） 0 moreover, after the joint application of 10- 15 μg/ml triptolide and 1μg/ml cisplatin, the inhibition rate became significantly higher than that of the respective application. Triptolide and cisplatin inhibited the MG63 cells respectively and jointly in a time-dependent manner. [Conclusion] Triptolide and cisplatin can inhibit the proliferation of osteosarcoma cells respectively or jointly.
