摘  要:以小黑杨为受体材料,在对传统的农杆菌介导的叶盘转化法改进的基础上进行了转化外源几丁质酶基因。试验表明,菌液浓度为0.3~0.4是小黑杨遗传转化的适宜浓度,0.3时分化率最高(24.75)。菌液浓度超过0.45时,分化率逐渐下降。经过对转化子的PCR及PCR-Sothern鉴定,结果呈阳性,证明外源几丁质酶基因导入并整合进小黑杨基因组中。Populus simonii × p. nigra was served as the acceptor . The foreign chitinase gene was introduced by modified agrobacterium tumefaciens mediated method into Populus simonii x p. nigra. The result showed that 0.3-0.4 was the suitable concentration of bacterium liquid, 0.3 has the highest transformation ratio (24.75) . The differentiation rate was descended when bacterium liquid was 0.45, transformation plants was tested by PCR and PCR Southern, the result is positive, it is showed that the foreign chitinase gene was already introduced into genome of Populus simonii× p. nigra.
