水杨酸模型大鼠杏仁核兴奋对下丘神经元放电的影响

摘  要:目的探讨边缘系统兴奋对耳鸣状态下听觉传导主要核团下丘外核(external nucleus nf inferiorcolliculus,ICx)放电的影响。方法采用细胞外记录方法,观察急性水杨酸(sodium salicylate,SA)耳鸣模型大鼠杏仁外侧核(lateral amygdaloid nucleus,LA)自发放电及电刺激 LA 后 ICx 自发放电活动和放电间隔直方图。结果1.SA 模型大鼠 LA 神经元平均自发放电率较正常对照组增加(5.42±0.68 Hz vs 3.36±0.24 Hz,P<0.01)。2.电刺激 LA 2h 后,正常对照组大鼠的 ICx 平均自发放电率较刺激前增加(7.90±0.85 Hz vs 3.21±0.41 Hz.P<0.01);0~50 ms 间隔放电脉冲比例由31.9%增加到55.1%。3.急性 SA 模型大鼠在电刺激 LA 前,ICx 的平均自发放电率(7.63±0.70 Hz)已高于正常对照组,其中0~6 ms 短间隔放电比例明显增加;刺激 LA 后2h 内,IGx 神经元平均自发放电率降低(5.66±1.05 Hz)与电刺激前比有差异(P<0.05),但较正常组刺激前,自发放电活动仍明显增加;0~20 ms 的 ICx 自发放电脉冲数比例明显下降(28.4% vs 14.5%)。结论大剂量 SA 可引起 LA和 ICx 神经元放电增加;SA 引起的 LA 神经元兴奋可引起 ICx 兴奋程度的改变,耳鸣是否伴随情绪因素,有待进一步研究。Purpose To explore the effects of excitation of the amygdaloid nucleus on the spontaneous discharge of external nucleus of inferior colliculus (ICx)under the tinnitus condition. Methods Extracellular microelectrode technique was used. Rat model for tinnitus was made by sodium salicylate (SA) injection. The spontaneous spikes of the lateral amygdaloid nucleus (LA), and that of ICx while LA was stimulated were observed. The interspike histograms were also recorded. Results 1. The average rate of spontaneous discharge recorded from LA increased significantly in SA treated group than that of control group(5.42± 0.68 Hz vs 3.36± 0.24 Hz, P 〈 0.01). 2. Two hours after LA electro-stimulation of LA, both the average rate of spontaneous discharge and short 0 - 50 ms interval spikes of ICx in the control groups increased significantly (7.90 ± 0.85 Hz vs 3.21 ± 0.41 Hz, P 〈 0.01 ) ( 31.9% to 55.1% ). 3. The average rate of spontaneous discharge of ICx in SA group(7.63 ± 0.70 Hz)was higher than that of the control group before LA stimulation. Two hours after stimulation of LA, the spontaneous discharge rate of SA model group decreased to 5.66 ± 1.05 Hz, which was still higher than that of the control group before stimulation of LA, and the percentage of 0 - 20 ms interval spikes obviously decreased from 28.4% to 14.5% .Condusion It is assumed that the increased spontaneous activity of ICx induced by salicylate is involved tinnitus. Excitation of the amygdaloid nucleus induced by salicylate affects ICx spontaneous activity in different way. These may be correlated with timfiuts accompanied with emotional component.
