摘 要:目的探讨以壳聚糖为佐剂的Hp疫苗的免疫保护作用及其机理。方法BALB/c小鼠随机分为7组空白对照组、壳聚糖酸溶液组、壳聚糖颗粒组、Hp抗原组、Hp抗原+壳聚糖酸溶液组、Hp抗原+壳聚糖颗粒组和Hp抗原+CT组,各组于第0、7、14和21天灌胃各免疫1次,末次免疫后4周给予SS1Hp菌攻击,隔日1次,共2次。在攻击前后分批处死小鼠,取胃黏膜检测Hp和Th1、Th2细胞因子含量,同时检测血清中抗HpIgG2a和IgG1含量。结果①以壳聚糖为佐剂的Hp疫苗的免疫保护率达60%,与以CT为佐剂的Hp疫苗的免疫保护率(58.33%)相似,显著高于单纯Hp抗原组及其他不含Hp抗原组(P<0.001~0.05)。②Hp攻击后胃黏膜内IFN-γ、IL-2和IL-12含量在含佐剂组显著高于无抗原和无佐剂组(P<0.001~0.05);③Hp攻击后胃黏膜内IL-4含量在以壳聚糖颗粒为佐剂组显著高于以CT为佐剂组(P<0.05),以壳聚糖溶液为佐剂组显著高于对照组、无佐剂组及佐剂中含CT组(P<0.001~0.05)。结论以壳聚糖为佐剂的Hp疫苗对Hp感染具有免疫保护作用,同时可促进Th1和Th2的混合免疫反应。Objective:To study the immunological protection of Hp vaccine with chitosa as adjuvant and it' s mechanism. Methods:One-grade female BALB/c mice were randomly divided into seven groups and immunized by PBS alone, chitosan solution alone, chitosan particles alone, Hp antigen alone, Hp antigen plus CT, Hp antigen plus chitosan solution and Hp antigen plus chitosan particles orally respectively once a week for four weeks. At 4 weeks after the last immunization, these mice were challenged by alive Hp twice at two days intervals. Mice were sacrificed at before and after challenge and gastric mucosa were embedded in paraffin, sectioned and assayed with Giemsa stain. The other gastric mucosa were used to quantitatively culture Hp. ELISA was used to detect IL-2, IFN-γ, IL-12, IL-4, IL-10 content in gastric mucosa, and to detect anti-Hp IgG1, IgG2a levels in serum. Results:①In the groups with chitosan as adjuvant, 60% mice could achieve immunological protection, which was according to that with CT as adjuvant(58. 33% ), and was significantly higher than Hp antigen alone and other groups without Hp antigen( P 〈0. 001 -0. 05 ). ②Aftcr challenge, the content of IFN-γ, IL-2 and IL-12 was significantly higher in the groups with adjuvant than those in the groups without adjuvant and antigen( P 〈 0. 001 -0. 05 ).③After challenge, the content of IL-4 was significantly higher in the groups with chitosan particles as an adjuvant than that in the group with CT as an adjuvant( P 〈 0. 05 ) , and that in the group with chitosan solution as an adjuvant was significantly higher than that in control group, non-adjuvant group and the groups with CT( P 〈 0. 001 - 0. 05 ). Conclusion :Hp vaccine with chitosan as adjuvant can protect against Hp infection, and may induce both Th1 and Th2 type immune response.
