摘 要:目的:研究9-芴甲氧羰基(Fmoc)固相合成法合成黑色素细胞刺激素(MSH)工艺的步骤及其影响因素.方法:采用Fmoc保护α-氨基,以TBTU/HOBT/D IEA为缩合剂合成MSH.用色谱仪和质谱仪对合成的多肽进行纯化、鉴定.结果:多肽合成的得率可达63%,经反相高效液相色谱仪(RP-HPLC)对合成的多肽进行纯化后可得纯度为98.67%的目标肽.通过基质辅助激光解吸附电离飞行质谱仪(MALD I-MS)检测所合成多肽的分子质量为1663.88 ku,与其理论分子质量1664.08 ku相符.结论:该合成法具有快捷、简便、高效地合成多肽MSH的特点,适合大批量生产.AIM: To study the process and influencing factors of synthesizing the melanocyte stimulating hormone (MSH) peptide by Fmoc solid-phase synthesis. METHODS: With Fmoc to protect the α-amino acid, TBTU/HOBT/DIEA were used as coupling reagents to synthesize the peptide of MSH. The synthesized peptide was purified by the chromatograph and identified by mass spectrograph. RESULTS: The peptide yield was 63%. The synthesized peptide purity reached to 98.67% after being purified by RP-HPLC. The molecular mass of the synthesized peptide was 1663.88 ku by MALDI-MS and it was consistent with the theoretic molecular mass of 1664.08 ku. CONCLUTION : This method of synthesis is rapid,simple and effective for the peptide MSH, and it's fit for the large-scale production.
