摘 要:目的:观察惊恐应激大鼠下丘脑单胺类神经递质及其代谢产物的变化,以及补肾固涩方药桑螵蛸散对其干预作用。方法:根据Takash i方法建立惊恐应激模型,采用HPLC-ECD检测惊恐应激后大鼠下丘脑单胺类神经递质及其代谢产物的含量,并与正常组、桑螵散给药组进行对照。结果:惊恐模型组大鼠下丘脑E、5-HT的含量显著下降,与空白对照组比较,差异显著(P<0.01,P<0.05);桑螵散给药组大鼠下丘脑E及5-HT含量、5-HT/DA比值明显升高,与模型组相比有显著性差异(P<0.05);模型组与空白对照组大鼠NE/DA、5-HT/DA、5-HT/NE比值无显著性差异(P>0.05)。结论:惊恐应激可以导致大鼠脏腑气机紊乱,下丘脑中E、5-HT含量下降可能是其发病机制之一;桑螵蛸散能改善大鼠惊恐状态,与调节下丘脑中E、5-HT含量和5-HT/DA比值有关。Objective: To investigate the changes of monoamine neurotransmittcr and their metabolite in hypothalamus of model rats with fear stress and the effects of Sangpiaoxiao Decoction(SPXD) Methods :To establish the fear stress model according to Takashi's method and determine the contents of monoamine neurotransmitters and their metabolites by HPLC - ECD. Compare fear group, SPXD group and normal group. Results : The lower contents of E, 5 - HT had been observed in hypothalamus of model rats compared with - normal group( P 〈 0.01, P 〈 0.05). On the contrary, the contents of both rose significantly in SPXD group. The comparisons of NE/DA , 5 - HT/DA and 5 - HT/NE in fear group and normal group have no significant discrepancy( P 〉 0.05 ) ,SPXD can regulate the 5 - HT/DA significantly ( P 〈 0. 05 ). Conclusion : The mechanism of Qi injured by fear sress is involved with the lower contents of E,5 -HT in hypothalamus,SPXD can improve the situation of fear by regulating the contents of E, 5 - HT and 5 - HT/ DA.
