摘 要:目的研究P-糖蛋白(PGP)和多药耐药相关蛋白1(MRP1)在脑室内注射红藻氨酸(KA)癫动物模型皮质内的表达。方法脑室内注射KA6、24h及1周后处死动物,采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学方法,观察PGP和MRP1在皮质的表达。结果KA注射24h后PGPmRNA(1.58±0.14)和免疫阳性细胞(19.36±1.64)在皮质表达显著增加,与空白对照组(mRNA0.73±0.06,免疫阳性细胞8.21±0.79)比较差异有统计学意义(P<0.05),1周后PGPmRNA(0.92±0.09)和免疫阳性细胞(8.42±0.84)表达下降至对照组水平;注射KA后MRP1表达逐渐升高,24h后MRP1mRNA(1.72±0.15)和免疫阳性细胞(26.36±2.32)表达显著增加,与空白对照组(mRNA0.87±0.07,免疫阳性细胞10.88±0.96)比较差异有统计学意义(P<0.05),1周后MRP1mRNA(1.57±0.12)和免疫阳性细胞(24.51±2.27)表达仍高于空白对照组(P<0.05)。结论KA诱发的癫动物模型皮质内PGP、MRP1表达增强,可能参与了癫耐药的发生。Objective To investigate the expression of PGP and MRP1 in cortex of epileptic model made by intraeerebroventricular injection of kainic acid (KA). Methods After intracerebroventricular injection of KA 6, 24 hours and 1 week, rats were respectively sacrificed and the brain were sampled. Then the expressions of PGP and MRP1 were observed in cortex in brain using RT-PCR and immunohistochemistry. Results Compared with those of control groups (mRNA 0. 73 ± 0. 06, immunoreaetive cells 8. 21 ±0. 79), PGP mRNA ( 1.58 ±0. 14) and immunoreactive cells ( 19. 36 ± 1.64) were significantly increased in cortex 24 hours after intracerebroventricular injection of KA ( P 〈 0. 05 ), and returned to normal level 1 week after KA injection ( mRNA 0. 92 ± 0. 09, immunoreactive cells 8. 42 ± 0. 84) ; Compared with those of control groups (mRNA 0. 87 ± 0. 07, immunoreactive cells 10. 88 ± 0. 96), MRPI mRNA ( 1.72 ± 0. 15 ) and immunoreactive cells ( 26. 36 ± 2. 32 ) showed an obviously statistical increasing in cortex 24 hours after intracerebroventricular injection of KA ( P 〈 0. 05 ), and still showed enhanced expression 1 week after KA injection (mRNA 1.57 ±0. 12, immunoreactive cells 24. 51 ±2. 27, P 〈 0. 05 ).
