摘  要:目的研究神经营养素-3(NT-3)对大鼠脊髓损伤后神经元中丝蛋白(NF200)及星形胶质细胞胶质纤维酸性蛋白(GFAP)表达的影响,探讨NT-3在脊髓损伤修复中的作用。方法SD大鼠24只,分为假手术组、损伤对照组和NT-3组,每组8只,于损伤前和损伤后1 d、7 d、14 d、21 d和28 d用BBB评分评定大鼠运动功能,然后处死动物用免疫组化染色及图像分析方法观察NF200及星形胶质细胞GFAP的表达。结果BBB评分,14 d、21 d和28 d点NT-3组明显高于损伤对照组;脊髓损伤后NF200及GFAP表达增加,且NT-3干预后表达更为明显,与其他2组相比差异有统计学意义(P<0.05)。结论NT-3促进星形胶质细胞增生及神经元功能的活跃,可能是促进神经纤维再生的一个重要因素。Objective To study the effects of neurotrophin-3 on NF200 and GFAP expressions in the injured spinal cord. Methods 24 SD rats were randomly divided into three groups: (1) sham operated group( n =8), (2)control group( n =8), and (3)NT-3 group( n = 8). Behavior test was carried out on the 8 rats from each group at 1 hour before spinal cord surgery, lday, 7 days, 14 days, 21days, and 28days after surgery using the BBB( Basso, Beattle, Bresnahan) scores. The immunohistochemical technique and image analysis were used to examine the expressions of NF200 and GFAP. Results The BBB scores were higher in the NT-3 group than in the control group at the time points of 2w, 3w and 4w; Both of NF200 and GFAP expressions were increased significantly after spinal cord injury, and the expressions in NT-3 group were significantly higher than those in the other two groups ( P 〈 0. 05 ). Conclusion NT-3 may play an important role in promoting the nerve regeneration by stimulating the hyperplasia of the astrocytes and the function of the neurons.
