急性细菌性角膜炎角膜中HF-κB的活化及二硫代氨基甲酸吡咯烷的保护作用

摘  要:目的探讨核转录因子-kB(NF-κB)及细胞间粘附分子-1(ICAM-1)在急性细菌性角膜炎大鼠角膜中的表达及二硫代氨基甲酸吡咯烷(PDTC)对两者表达的影响。方法用绿脓杆菌脂多糖(LPS)建立急性细菌性角膜炎大鼠模型,模型制作前30min将生理盐水和PDTC分别注射于大鼠球结膜下(分别为炎症组;PDTC预处理组),在模型制作后0.5h、1h、3h、6h、12h、24h、及72h时,分别用裂隙灯显微镜观察大鼠眼部表现并给予评分;免疫组织化学法检测大鼠角膜组织中NF-κB的活化情况;逆转录聚合酶链反应(RT-PCR)检测大鼠角膜组织内ICAM-1mRNAR的表达。结果应用PDTC预处理后,PDTC预处理组大鼠的角膜炎症状、角膜组织的损害均较炎症组明显减轻;ICAM-1mRNA的表达也较炎症组明显降低,免疫组组织化学染色显示PDTC预处理组的NF-κB阳性细胞数明显少于炎症组。结论NF-κB及其诱导的ICAM-1在急性细菌性角膜炎的发病过程中发挥重要作用;PDTC可有效减轻角膜炎症,此作用可能与其抑制NF-κB活化,进而减少细胞因子产生有关。Objective To investigate the expression of nuclear factor- κ B(NF- κB) and intercellular adhesion molecule- 1 (ICAM- 1) in corneas of acute bacterial keratitis in Wistarrats, and the effect of pyrrolidine dithiocarbamate(PDTC) on the expression of NG- κ B and ICAM-1. Methods Pseudomonas aeruginosa Lipopolysaccharide (LPS) -induced keratitis models were established in wistar rats.Before LPS exposure 30 minutes,PDTC and normal saline were respectively injected into subconjunctival tissues of rats.At 0.5,1,3,6,12, 24 and 72 hours after LPS exposure, each rat's clinical representation was observed with silt-lamp microscope. Then rats were killed and corneas were excised.The activation and distribution of NF- κ B were evaluated with Immunohistochemical analysis,the expression of ICAM-1 reRAN in the cornea was detected by reverse transcription-polymerase chain reaction(RT-PCR). Results Compared to the rats of LPS group, both symptoms and lesions of cornea were slighter in the rats of PDTC pretreated group. Furthermore, the expression of ICAM- 1 was significantly decreased and the activation of NF- κ B was also suppressed in PDTC pretrated group as compared with LPS group. Conclusion NF- κ B and ICAM-1 may play an important role in bacterial keratitis, as the inhibitor of NF-κ B, PDTC could deduce the corneal inflammation,which might be due to inhibition of NF- κ B activation and down-regulation of ICAM-1 m RNA.
