兔高同型半胱氨酸血症模型建立及动脉粥样硬化形成的实验研究

摘　　要：[目的]观察同型半胱氨酸(homocysteine,Hcy)致兔高同型半胱氨酸血症模型建立的剂量、时间及动脉粥样硬化形成的特点。[方法]高、低剂量组分别皮下注射Hcy 20mg/(kg.d)、15mg/(kg.d),并设生理盐水组为对照。分析各组注射前、注前后4周及11周血清中Hcy值,采用B超观察动脉狭窄程度及病理切片观察动脉粥样硬化形成的情况,实验结束分别测定各组血清中TG、TC值。[结果]皮下注射Hcy 4周时血中Hcy明显高于对照组(P<0.01),髂总动脉出现狭窄,11周时颈总动脉、髂总动脉及其分支有明显的动脉粥样硬化征象,部分主动脉也有动脉粥样硬化形成。[结论]予15mg/(kg.d)皮下注射Hcy 4周可建立高同型半胱氨酸血症模型,11周时可形成动脉粥样硬化,其所致的动脉粥样硬化首先在小动脉形成,提示同型半胱氨酸是动脉粥样硬化形成的一个独立危险因素。[Objective]To calculate the dosage of homocysteine（ Hey） used to induce the rabbit homocystinemia model, the time involved, and to observe the characteristic of Artherosclerosis formation. [ Methods]Rabbits were randomly divided into 3 groups： the high dosage group, the low dosage group and the control group. The first two groups were injected hypodermically with Hcy 20mg/[kg·d] and 15mg/[kg·d） respectively, while the control group was given normal saline. The levels of Hey in serum in each group were analyzed before injection, and 4 weeks and 11 weeks after injection. The arterial stenosis was investigated by type-B ultrasonic and the formation of artherosclerosis was observed through pathological section. Moreover, the levels of TG, TC in serum of each group were determined. [Results]After 4 weeks of injection, the levels of Hcy in serum in the two dosage groups were significantly higher than the control group （ P 〈 0.01） and stenosis appeared in common iliac artery. 11 weeks later, the indicator of artherosderosis was seen in the main arteries, especially obvious in common carotid artery, common iliac artery and its branches. [Conclusions] When administrated with Hey injection [15mg/（kg·d） ], rabbit can be made into the homocytinemia model in the 4th weeks and artherosclerosis can be developed in the 11th week with the first affected arteries being the arterioles. It is suggested Hey is an independent risk factor for the formation of artherosclerosis.
