摘  要:目的:用不同的方法制备Hsp70-HSV-2gD融合蛋白疫苗阳离子脂质体,探索Hsp70-HSV-2gD融合蛋白疫苗可行的应用途径。方法:采用冻融法、逆相蒸发法、钙融合法分别制备Hsp70-HSV-2gD融合蛋白疫苗阳离子脂质体,在透射电镜下观察脂质体颗粒的大小,应用紫外分光光度计检测蛋白疫苗的包封率,并用ELISA法对Hsp70-HSV-2gD抗原活性进行测定。结果:逆相蒸发法制备的Hsp70-HSV-2gD融合蛋白疫苗阳离子脂质体包封率最大,达到(53±2.0)%,所制备的脂质体粒圆形,直径大小较一致,约600 nm,具有抗原活性。结论:成功制备了Hsp70-HSV-2gD融合蛋白疫苗阳离子脂质体,逆相蒸发法为制备Hsp70-HSV-2gD融合蛋白疫苗阳离子脂质体的最好方法。Objective: To study the different methods of preparing Hsp70 - HSV - 2gD fusion protein vaccine cationic liposomes and to explore the possible approach in application. Methods: Liposomes were made by freezethawing methods, reverse phase evaporation methods and calcium- induced fusion methods. The shape and size of the liposomes granule were observed by transmission electron microscope and the encapsulation efficiency of the liposome was detected by spectrophotometer. The antigen activity of Hsp70- HSV - 2gD fusion protein was tested by ELISA. Results: Encapsulation efficiency of Hsp70- HSV - 2gD fusion protein vaccine cationic liposomes made by reverse phase evaporation methods was the best, being 53± 2.0%. The liposomes were rotmd and the diameter of granules was unanimous, and the average size of the complexes was 600nm, with antigen activity. Conclusion: The Hsp70 - HSV - 2gD fusion protein vaccine cationic lipusomes is prepared successfully and reverse phase evaporation method is the best way to prepare these hposomes.
