脂糖舒对糖尿病大鼠内脏器官结构损伤的保护作用

摘  要:目的探讨脂糖舒对糖尿病大鼠内脏器官结构损伤的保护作用。方法腹腔注射链脲霉素制作糖尿病大鼠模型。用不同剂量脂糖舒(0.5g/kg和1.0g/kg)灌胃,连续8周,并取健康大鼠作正常对照。采用病理学检验技术观察各组大鼠心肌、肾脏和肝脏等器官的病理学变化,并计算出心肌、肾脏和肝脏的器官系数。结果模型组心肌纤维水变性,结构松散,胞浆内见较多细小颗粒。肾小球毛细血管轻度增生,呈分叶状。部分肾小管上皮细胞水肿,管腔内可见蛋白样渗出物。肝小叶结构不清,肝索、肝窦比例失调,肝细胞胞浆疏松化,部分区域肝细胞索解离,细胞核固缩,核碎裂,核溶解,灶性坏死。肝和肾的器官系数较正常组差异显著(P<0.01)。而经脂糖舒处理后,大剂量组对糖尿病大鼠肾脏系数较模型组明显降低(P<0.01),对心、肾和肝脏结构损伤的改善作用较其小剂量组明显。结论脂糖舒对糖尿病大鼠内脏结构损伤有明显的保护作用。Objective To Observe the protective effect of Zhitangshu (ZTS) on the visceral organss structure injury of diabetic rats. Methods The diabetic model of rats was established by abdominal injection of steptozotocin(50mg/kg). The different doses of ZTS (0.5g/kg and 1.0g/kg) were respectively perfused into stomach for 8 weeks. And other healthy rats were used as control. The pathologic changes of visceral organs' structures(heart, kidney and liver) of rats in each group were respectively examined by using histopathological technique,and the coefficient of these Organs were counted. Results In model group; there were vesicular degeneration in myocardial cells, its structures showed loosen and a lot of particles appeared in cytoplasm;the glomeruli capillaries lightly proliferated and became lobular; some of tubular epitheliar cells were swelling and there were proteinoid exudate in these tubular cavities;the lobuar structures in liver were cloudy, the proportion of hepatocyte cord and sinus was disordered. The endolysis in hepatocytes was showed and partial hepatocyte cord were resolved, some of hepatocytes showed nucreo-pycnosis, even-lysis and the spot-necrosis could be seen under Optical microscope(OM), There were significant difference in coefficients of kidney and liver between diabetic control group and normal group( P〈0.01). By using ZTS, the coefficients of kidney in the high doses of ZTS group was clearly lowered (compared with diabetic control group P〈0.01). The improve effect of the high doses of ZTS on the Structure injury of myocardium,liver and kidney in diabetic rats were obviously showed under OM. Conclusion ZTS has obvious protective effect on visceral structure injury of diabetic rats
