摘 要:目的探讨山楂叶总黄酮(TFHL)对小鼠的抗氧化作用和体外清除羟自由基(OH-)的作用。方法灌胃给予7月龄小鼠TFHL,连续30d后,采用生化方法测定小鼠全血的超氧化物歧化酶(SOD)和谷胱甘肽过氧物酶(GSH-Px)活性,并检测小鼠心、肝、脑组织中脂褐质(LF)、丙二醛(MDA)和SOD含量。采用体外抗坏血酸体系产生OH-的原理,观察TFHL清除OH-的作用。结果给予TFHL(40mg/kg,80mg/kg)的小鼠心、肝、脑组织中SOD含量和全血中SOD和GSH-Px活力显著提高,而组织中MDA和LF含量则明显下降。TFHL亦能清除抗坏血酸体系产生的OH-,且OH-的清除率呈明显的量效相关性(r=0.995)。结论TFHL具有直接清除机体OH-,提高组织中抗氧化酶的活性,抑制自由基损伤,防止脂质过氧化的作用。Objective To study the antioxidant effect of total flavone of hawthorn leaf (TFHL) in mouse and its effect of scavenging hydroxyl free radical ( OH^- ) in vitro. Methods The mice were treated with TFHL (40 mg/kg, 80 mg/kg) through oral gavage daily for 30 d, and to determine the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in blood and the contents of lipofuscin (LF), malondialdehyde (MDA) and SOD in liver, brain and heart by biochemical methods. OH was induced by ascorbate-Cu^2+ reaction system, and to estimate the effect of TFHL on scavenging OH by colorimetry. Results In TFHL group the activities of SOD and GSH-Px in blood and the activities of SOD in liver, brain and heart were significantly increased, and the contents of LF and MDA in mice tissues were obriously reduced TFHL from 12.5 mg/L to 150 mg/L had marked effects on scavenging OH from 38.5% -100%,and it showed dose-relationship (r=0. 995). Conculsion TFHL can obviously inhibit damage of free radical and prevent lipid peroxidation, the mechanisms maybe associated with directly scavenging the OH and increasing the activities of antioxidation enzymes in mice tissues.
