摘 要:采用微吸管实验技术将研究层粘连蛋白(Laminin,LN)(LN)作包被液预裱衬处理微吸管后,LN趋化液对了解探讨肝癌(HCC)细胞响应LN时的趋触效应趋化作用以及基质在对微吸管表面的粘附的作用对肝癌细胞(HCC)以及肿瘤细胞伪足形成的影响。研究不同浓度LN趋化液对HCC细胞与不同浓度LN的趋化作用。定量描述肝癌细胞(HCC)在层粘连蛋白LN(Laminin,LN)趋化作用下伪足的形成、生长的动态过程。结果显示,当微吸管未经LN预裱衬处理,而仅放置空白趋化液时未见有伪足形成;微吸管内壁经100μg/mlLN预裱衬后再加入空白趋化液时,仍未见伪足形成;对微吸管内表面与未经LN预裱衬处理或比较,微吸管中经加入50μg/ml LN趋化液预裱衬处理微管内加入50μg/ml LN趋化液时肝癌细胞所形成的伪足峰值长度的比较,在两种情况下,二者无显著性差异(P>0.05)。当LN趋化液浓度趋化剂LN浓度≤200μg/ml,HCC细胞伪足峰值的生长长度随浓度的增加而增大加;当LN浓度≥200μg/ml,伪足的生长呈现饱和性。提示微吸管内表面裱衬LN对肝癌细胞伪足形成的趋化作用可忽略。在HCC细胞响应LN趋化作用生成伪足具有一定范围的浓度依赖性和时间依赖性。LN在一定浓度范围内,对HCC细胞的趋化作用具有浓度依赖性。A micropipette thchnique was adopted to investigate hepatocelluiar carcinoma cells (HCC)cells in response to chema haptotaxis taxis of laminin (LN)onto LN coated surfaces and effect of LN adhesion to pseudopod protrusion of HCC cells on the surface of micropipette.And ctne formation & dynamical growth hemotaxis of pseudopod protrusion of HCC cells in response to a variable concentration of LN stimulation was expressdemonstrated.T quantitativeiy describe the formation & dynamical growth of hepatocelular carcnoma (HCC)cells to laminin(LN)chemotaxis.The results showed that Cchemotactic pseudopod protrusion of a HCC cel was unvisible filing with bland solution into a micropipette, which wasn't without precoated LN.And pseudopod protrusion wasn't still observed when a micropipette coated surtace with LN of 100μg/ml was filed in bland solution with precoated LN of 100 μg/ml/No matter what any treatment coated or not noto the surface of micropipette with LN was adopted and LN was filled in pipette with LN of 50 μg/ml in tow cases(P〉0.05).The length of pseudopod protrusion of HCC cells was viewed,which wasn't notable different between tow cases.Along the increment of concentration within LN of 200 μg/ml,the length of chemotactic pseudopod protrusion of a HCC cell would increase,But it attained seturated length up to LN of 200 μμg/ml.The results suggests that the factor of haptotaxis can could be neglected while studying on HCC cells in response to LN chemotaxis with a singlemicropipette.And strength of chemotaxis dependss on the increment of concentration within some concentration and chemotactic duration of LN.
