摘 要:探讨了木素在黄柳(Salix gordejecii)纤维细胞壁中的分布情况及其与制浆造纸的关系.采用透射电子显微镜(TEM)、扫描电子显微镜结合X射线能谱仪(SEM-EDXA)及共聚焦激光扫描显微镜(CLSM)研究了黄柳纤维细胞壁的分层结构,以及木素在细胞壁各层中的分布.TEM结果表明,黄柳纤维细胞壁分为胞间层、初生壁、次生壁外层、次生壁中层及次生壁内层.CLSM图像显示木素在纤维细胞各个壁层中分布不均一,细胞角隅区木素含量最高、其次是胞间层,次生壁最低.导管中的木素浓度比纤维细胞中的高.SEM-EDXA研究表明细胞角隅区、胞间层、次生壁中层的木素浓度比为2.15∶1.32∶1.The distribution of hgnin in the cell wall of Salix gordejecii fiber and its effect on pulping and papermaking were studied. The uhrastructure of the cell wall of Salix gordejecii fiber and the distribution of lignin in the cell wall were then determined using TEM, scanning electron microscopy combined with energy dispersive X-ray analysis (SEM-EDXA) and confocal laser scanning microscopy (CLSM). The TEM images show that the cell wall of Salix gordejecii fiber is typically divided into three layers, namely the middle lamellar, the primary wall and the secondary wall that includes the outer, the middle and the inner layers, The CLSM images reveal that the distribution of lignin in the cell wall layers is not uniform, following a decreasing order of the cell comer, the middle lamellar and the secondary wall, and that the lignin concentration in the vessel is larger than that in the fiber cell. Moreover, the SEM-EDXA results indicate that the lignin concentration in the cell comer, the middle lamellar and the middle layer of the secondary wall is in a ratio of 2.15: 1.32: 1.
