摘  要:目的:观察维生素C对糖尿病大鼠血管内皮主动脉细胞间黏附分子1 mRNA表达的调控作用,以及对血脂、血糖水平的影响。方法:实验于2005-06/12在南华大学附属第一医院临床医学研究所和娄底市中心医院药剂科完成。①选用成年雄性SD大鼠40只。取大鼠30只,利用链脲佐菌素腹腔注射法诱导建立1型糖尿病大鼠模型,造模成功20只,采用随机区组法将其分为2组:治疗组(n=10):按100mg/(kg·d)灌胃1g/L维生素C(天津药业焦作有限公司,批号:040217-1),1次/d;模型组(n=10):灌胃等量生理盐水。其余10只大鼠设为对照组:注射等量柠檬酸钠缓冲液。每组干预16周。②干预结束后,用全自动生化仪测大鼠血清空腹血糖、糖化血红蛋白、三酰甘油、总胆固醇、低密度脂蛋白胆固醇、高密度脂蛋白胆固醇,并测定血清糖化低密度脂蛋白水平。分离主动脉,原位杂交检测主动脉内膜细胞间黏附分子1 mRNA表达。③均数间比较采用单因素方差分析,组间差异比较采用t检验。结果:因造模失败脱失10只,最终进入结果分析30只。①主动脉组织细胞间黏附分子1 mRNA表达:模型组与对照组比较,主动脉内膜层细胞胞浆细胞间黏附分子1 mRNA表达显著上调,分别为66.38±8.69,29.33±4.39(P<0.05);治疗组与模型组比较,细胞间黏附分子1 mRNA阳性表达减弱(49.78±8.63,P<0.05)。②血糖水平:干预后模型组明显高于对照组(P<0.05),治疗组与模型组相近(P>0.05)。③血脂水平:干预后模型组血清三酰甘油、总胆固醇、低密度脂蛋白胆固醇、糖化血红蛋白、糖化低密度脂蛋白水平明显高于对照组,高密度脂蛋白胆固醇水平明显低于对照组(P<0.05)。干预后治疗组血清三酰甘油、总胆固醇、低密度脂蛋白胆固醇、糖化血红蛋白、糖化低密度脂蛋白水平明显低于模型组,高密度脂蛋白胆固醇水平明显高于模型组(P<0.05)。结论:维生素可下调主动脉组�AIM: To observe the regulation effect of Vitamin C on the expression of intercellular adhesion molecule-1 (ICAM-1) in the aorta of blood vessel endothelium (BVE) of rats with diabetes as well as its effects on the levels of blood lipid and blood glucose. METHODS: The experiment was conducted in the Department of Clinical Medicine, First Hospital Affiliated to Nanhua University between June and December 2005. ① Forty male adult SD rats were selected. Thirty rats ware induced into models of type I diabetes by peritoneal injection of streptozotocin (STZ) and 20 successful models were randomly divided into 2 groups. Rats in the treatment group (n =10) ware administrated by gastric perfusion of Vitamin C (provided by Tianjin Jiaozuo Pharmaceutical Co., Ltd with the batch number of 040217-1) once a day at 1 g/L according to 100 mg/kg per day. Rats In the model group (n =10) were administrated by gastric perfusion of normal saline at the same dose. Other 10 rats ware taken as the control group and were injected with sodium citrate at the same dose. Rats in each group ware interfered for 16 weeks. ② The serum fasting blood sugar, glycosylated hemoglobin, triglYceride, total cholesterol (TC), low density lipoprotein-cholesterol (LDL-C), high density lipoprotein-cholesterol (HDL-C) and the level of serum glycosylated low density lipoprotein (LDL) ware determined with automatic biochemical analyzer after the intervention. The aorta was separated and the expression of ICAM-1 mRNA in the BVE aorta was detected with in situ hybridization. ③ Differences among means ware compared with single factor analysis of variance, and differences among groups ware compared with t test. RESULTS: Ten rats failed in the modeling and withdrew from the study. Thirty rats finally entered the analysis of results. The expression of ICAM-1 mRNA in the BVE aorta: It was significantly up-regulated In the model group in comparison with that in the control group, which were 66.38±6.69,29.33±4
