平喘灵对哮喘大鼠IL-4mRNA和IFN-γmRNA表达及IgE含量的影响

摘  要:目的:探讨平喘灵对哮喘大鼠IL-4mRNA和IFN-γmRNA表达及IgE含量的影响。方法:SD大鼠分为空白对照组、模型组、平喘灵大、小剂量组(剂量分别为5.76,2.88g·kg^(-1))、地塞米松组(0.81mg·kg^(-1))和平喘灵小剂量(2.88g·kg^(-1))加地塞米松小剂量组(0.405mg·kg^(-1)),每组10只。用卵蛋白雾化吸入致敏法制备大鼠哮喘模型,于每天激发前12h分别灌胃相应剂量的药物,共7d。采用免疫放射法、逆转录聚合酶链反应(RT-PCR)半定量法分别检测各组大鼠血清和肺泡灌洗液(BALF)中的IgE含量以及肺组织IL-4 mRNA和IFN-γmRNA的表达量。结果:与空白对照组比较,模型组大鼠IgE含量、IL-4mRNA表达显著升高,IFN-γmRNA表达显著降低,IL-4/IFN-γ比值明显升高,且引喘潜伏期明显缩短,差异均有显著性(P<0.05)。与模型组比较,平喘灵组血清IgE的含量、IL- 4的表达明显降低,IFN-γ的表达明显升高,IL-4/IFN-γ比值明显降低;呼吸状态明显改善,且引喘潜伏期明显延长,均有统计学差异(P<0.05);地塞米松小剂量加平喘灵小剂量组对上述各项指标的改善作用较平喘灵小剂量组更明显,且在对血清及肺灌洗液IgE含量的调节方面也优于大剂量平喘灵。结论:平喘灵可能是通过调节IL-4/ IFN-γ平衡,继而调节IgE的产生。从而达到治疗哮喘的目的,且平喘灵与激素具有协同效应。Objective: To assess the modulation of levels of IL-4mRNA, IFN-γmRNA and IgE with Pingchuanling (PCL) in rats with bronchial asthma. Methods: The SD rats were randomly assigned into one of treatment groups: healthy normal control, asthma model control, dexamethasone dose (0. 81 mg·kg^-1), two PCL doses (5.76 and 2.88 g·kg^-1 ) or dexamethasone (0. 405 mg·kg^-1) plus PCL (2. 88 g·kg^-1). Each group had 10 SD rats. The asthma model of rats was induced by aerosolized ovalbumin inhalation, and the treatment groups were ig administrated for 7 days with corresponding drugs 12 hours ahead of provocation. Quantification of IgE in serum and bronchoalveolar lavage fluids (BALF) and IL-4 mRNA and IFN-γ mRNA levels in lung tissues were conducted by immuno-radiometric assay (IRMA) and RT-PCR. Results: Compared to the normal control,the model rats had signifi of serum IgE level and IL-4 mRNA expression, and a decrease of IFN-γ mRNA expression, higher IL-4/IFN-γ ratio and shorter asthma-induced delitescence ( P 〈 0. 05 ). Compared cant increases manifesting a to the model control, the PCL-treated rats showed significant reductions of serum IgE count and IL-4 mRNA expression, and escalation of IFN-γmRNA expression, displaying a reduced IL-4/IFN-γratio, improved respiration state and prolonged asthma-induced delitescence (P 〈 0. 05 ). The rats in the bi-therapeutic regimen were more evident in the improvements of symptoms and IL-4/IFN-γ index and better regulation of IgE count in serum and BALF than the rats receiving PCL alone. Conclusion: PCL relieved bronchial asthma by modulating IL-4/IFN-γ ration followed by IgE level and was synergistic with dexamethasone.
