摘 要:目的观察缬沙坦干预治疗后糖尿病大鼠肾脏nephrin表达的变化,探讨缬沙坦保护肾脏的部分机制。方法以链脲佐菌素建立糖尿病大鼠模型,缬沙坦干预治疗10周,观察大鼠糖代谢、肾功能及肾小球中nephrin表达的变化。结果模型组大鼠24 h尿蛋白含量显著增加,肾小球nephrin表达显著减少,而缬沙坦干预治疗组上述指标改善(P<0.05),且蛋白尿的程度与nephrin表达呈负相关(P<0.05)。结论缬沙坦可减少糖尿病肾病大鼠尿蛋白的排出,其机制可能与其减少肾小球nephrin表达有关。Objective To observe the protective effect of valsartan(an angiotensin receptor blocker) on the kidney function and nephrin expression in diabetic rats,and to study the possible mechanisms of valsartan protecting the kidney function of DN. Methods We induced the diabetic rat model by a single intraperitoneal injection of streptozocin(STZ). After 10 weeks treatment with valsartan, to assess the change of glucose metabolism, kidney function and nephrin. Results The test showed that the volume of 24 hour urine albumin in model group significantly increased and the expression of nephrin in kidney significantly decreased while it significantly ameliorated in valsartan group. Conclusion Valsartan decreased the urine albumin excretory rate in rats with diabetes meUitus nephrothy,which mechanism may be at least partly correlated with lessening the reduction of expression of nephrin in kidney.
