摘 要:目的:阐述钙离子与钙网织蛋白对心肌细胞的作用。资料来源:检索PubMed 1994-01/2005-12与钙离子和钙网织蛋白对心肌细胞作用的相关文章,检索词为“Ca2+,calreticulin,cardiomy-ocytes/cardiac myocyte,differentiation”,并限定语言种类为英语。资料选择:对所检索到的文献进行初筛,选择与钙离子和钙网织蛋白对心肌细胞作用的相关文章,筛除明显不符合原文的文章和重复性研究。资料提炼:共收集相关文章200篇,纳入42篇,排除158篇。资料综合:通过对内质网和肌质网上细胞内Ca2+释放通道(RyR和IP3R)的分子结构以及crt基因结构的研究表明:通道分子和钙网织蛋白共同参与调控体内Ca2+稳态,从而在心脏发育分化过程中发挥重要作用。结论:在心脏中细胞内游离Ca2+是一种关键的第二信使,细胞内Ca2+稳态的改变会显著影响许多心脏功能包括收缩舒张和心脏发育。细胞内Ca2+稳态的维持主要依赖内质网。在内质网中,钙离子主要与钙网织蛋白结合。crt基因敲除可损伤心脏发育而导致胚胎死亡。OBJECTIVE: To elucidate the important role of calcium and calreticulin in cardiomyocyte DATA SOURCES: We searched PubMed database for articles about the role of calcium and calreticulin in the cardiomyocyte published in English from January 1994 to December 2005 with the key words of "Ca^2+, calreticulin, cardiomyocytes/cardiac myocyte, differentiation". STUDY SELECTION: The retrieved data were selected primarily and articles about the important role of calcium and calreticulin in the cardiomyocyte were selected. Meanwhile, articles of repetitive research were excluded. DATA EXTRACTION : Totally 200 articles were researched, 42 were included, and 158 were excluded DATA SYNTHESIS: Investigation on the molecular structure of the intracellular Ca^2+ release channels (RyR and IP3R) on the endoplasmic reticulum and sarcoplasmic reticulum and structure of crt gene manifests that these channels co-participate in the regulation of calcium homeostasis with calreticulin, which plays a key role in cardiac development and differentiation. CONCLUSION : Intracellular free Ca^2+ is a critical second messenger in heart. Alterations in intracellular Ca^2+ homeostasis have profound effects on cardiac functions such as contraction-relaxation and cardiac development. Intracellular Ca^2+ homeostasis is maintained primarily via the endoplasmic reticulum, within which, Ca^2+ is mainly bound to calreticulin. Knockout of crt gene is embryonically lethal due to impaired cardiac development.
