高能量饮食雌性运动大鼠脂肪组织瘦素mRNA的表达

摘  要:目的:瘦素作为调节机体能量平衡的因子在生殖方面亦具有重要的调节作用,观察高能量饮食对雌性运动大鼠脂肪组织瘦素分泌的影响。方法:实验于2005-07/12在上海体育学院运动科学实验中心完成。选用32只SD雌性大鼠,随机(抽签法)分为4组,即正常组、高能量饮食组、运动组、运动+高能量饮食组,每组8只。正常组:常规基础饲料膳食。高能量饮食组:高能量饲料膳食(每200g基础饲料中加入鸡蛋15g,食糖40g,猪油20g,浓鱼肝油10滴)。运动组:采用递增负荷运动。运动+高能量饮食组:采用递增负荷运动,同时膳食采用高能量饲料。各组大鼠膳食平均40g/(d·只)。除对照组外运动组均采用段氏PT-98型进行运动训练,先适应性进行训练5d,以后每周运动6d,休息1d,训练2次/d,上下午各1次,最后1d的晚上加训1次。负荷设定参考Bedford的动物运动负荷标准,1~6周跑台速度分别为15,20,27,32,35,35m/min,40min/次,坡度为0°。所有组均于4周后隔天行阴道脱落细胞学检查,保证大鼠于动情间期处死,各组交叉进行。观察高能量饮食对雌性运动大鼠的体质量、体内脂肪含量及脂肪组织的瘦素mRNA表达的影响。结果:32只大鼠均进入结果分析。①各组大鼠体质量、腹内脂肪质量的比较:经过高能量饲料喂养后,高能量饮食组的体质量、腹腔内脂肪湿质量及单位体质量脂肪明显高于正常组[(323.58±23.88),(281.01±12.67)g;(11.53±2.99),(6.01±2.10)g;(35.35±7.24)×103,(18.62±2.89)×103;P<0.01];运动组各项指标均低于高能量饮食组(P<0.01);运动+高能量饮食组的体质量、腹内总脂肪湿质量及单位体质量脂肪均高于运动组,但差异无统计学意义(P>0.05)。②各组大鼠脂肪组织瘦素mRNA表达的比较:运动组与正常组相比脂肪组织中的瘦素mRNA表达水平明显下降(P<0.01);而运动+高能量饮食组脂肪组织中的瘦素mRNA表达水平明显高于运动组(P<0.01),运�AIM: As a kind of regulator in energy balance, leptin plays an important role in regulating the reproduction function. To observe the effect of the high-energy food on the secretion of leptin in adipose tissue of female athletic rats. METHODS: The experiment was conducted in the Experimental Center for Sports Science of Shanghai Institute of Physical Education from July to December 2005. Thirty-two female SD rats were selected and divided randomly into 4 groups: normal group, high-energy diet group, exercise group, exercise and high-energy diet group with 8 rats in each group. Rats in the normal group were fed with conventional food. Rats in the high-energy diet group were fed with high-energy food (200 g of food containing 15 g of egg, 40 g of sugar, 20 g of lard and 10 drops of oleum jecoris piscis concentratum). Rats in the exercise group were given increased loading training. Rats in the exercise and high-energy diet group were given progressive load exercise as well as high-energy diet simultaneously. Rats in each group were fed with averagely at 40g/day. Rats except those in the control group were trained to exercise on Duan PT-98 treadmill. Rats were trainedfor 5 days for adaptability, and then exercised 6 days a week with one day rest. The training was conducted twice a day respectively in the moming and aftemoon. Training in the aftemoon of the last day was doubled. The load was set down by referring to the standard of Bedford animal training, and rats ran on a treadmill of zero degree at the speed of 15,20,27,32,35,35 m/minute respectively from the 1^at to the 6^th week with 40 minutes each time. After 4 weeks, rats in all groups were examined of the desquamated vagina cells every other day to guarantee that rats were executed at anestrum, which was conducted in each group crossly. The effect of high-energy food on the body mass, adipose content in vivo and the expressions of leptin mRNA in adipose tissues of female athletic rats were observed.
