豆楮方对DEN诱发大鼠肝癌的防治作用

摘　　要：目的探讨中药复方豆楮方对DEN诱发大鼠肝癌的干预作用。方法50只SD大鼠在第1周按剂量为100 mg/kg注射DEN后,从第2周起按每周70 mg/kg剂量,连续灌胃16周诱发大鼠肝癌模型,用药组给予不同剂量的豆楮方灌胃,第18周处死大鼠,观察大鼠肝癌结节数、肝/体质量比、肝脏病理变化及bcl-2/bax免疫组化表达。结果豆楮方大鼠肝癌结节数、肝/体质量比、血清及肝组织匀浆的GST、血清ALT、AST水平较模型组有所降低,而豆楮方大鼠的A/G比例较模型组明显上升,豆楮方还能对抗DEN造成的大鼠肝脏bcl-2上调及bax下调。结论豆楮方具有防治DEN诱发大鼠肝癌的作用,其作用机制可能与基因调控有关。OBJECTIVE To investigate into the effect of compound Douehu Recipe in interfering with rat liver cancer induced by diethylinitrosamine （DEN）. METHOD Fifty SD rats were injected DEN at the dosage of 100 mg/kg in the first week and were injected DEN at the dosage of 70mg/kg for the following 16 weeks, starting from the second week, to create liver cancer models. The drug group was given different dosages of Douebu Recipe and killed in the 18th week. Observation was made on the number of liver cancer nodus, liver and body weight ratio, pathological changes of the liver and the expression of bcl-2/bax immunity group. RESULT The number of liver cancer nedus, liver and physique ratio, and the levels of GST and serum ALT and AST in the liver tissue homogenate in the drug group decreased as compared with the model group, but A/G ratio increased. Douehu Recipe could also antagonize bcl-s increase and bax decrease caused by DEN. CONCLUSION Douebu Recipe has the effect of preventing and treating rat liver cancer induced by DEN, the mechanism of which is probably related to the regulation of genes.
