摘 要:目的探讨小鼠脾脏CD8^+ T细胞的免疫磁珠负性分选方法,并对分选后所得细胞进行纯度、活力及功能检测。方法以免疫磁珠负性分选法从小鼠脾脏细胞中分离CD8^+ T细胞,流式细胞术检测所得细胞的纯度,台盼蓝检测细胞活力并用ConA刺激检测增殖能力。结果经过流式细胞仪测定免疫磁珠负性分选后的小鼠脾脏CD8^+ T细胞纯度达到(91.6±3.6)%,台盼兰染色细胞活力为(94.9±3.2)%,ConA刺激72h后有(56.3±1.7)%的细胞增殖。结论免疫磁珠负性分选法能够分选出高纯度的CD8^+ T细胞,并且不影响分选靶细胞的细胞活力和功能。Objective To assess the separation efficiency of magnetic-activated eel sorting (MACS) in the puxification of CD8^+ T cells from murine spleen. Methods CD8^+ T cells were isolated from murine spleen by MACS. Fluorescence-activated cell sorter was employed to determine the purity of CD^8 + T cells before and after the separation procedure. The viability and proliferation of the stored CD8^+ T cell were evaluated by trypan blue staining and ConA stimulation, respectively. Results The purity of CD8^ + T cells reached (91.6± 3.6)% after the purification with cell viability of ( 94.9 ± 3.2) % and proliferation of (56.3 ± 1.7) %. ,Conclusion CD8^ + T cells of high purity can be obtained from murine spleen using MACS without impairing the viability of the cells.
