定心胶囊对房颤大鼠P-选择素影响的实验研究

摘　　要：目的:探讨定心胶囊对房颤大鼠P-选择素的作用机制,研究开发中药治疗房颤及预防房颤后血栓。方法:采用氯化钙—乙酰胆碱混合液以鼠尾静脉给药的方式建立房颤的动物模型。筛选具有典型房颤心电图表现的大鼠30只,随机分为房颤模型组、定心胶囊治疗组、维拉帕米对照组和正常对照组,共4组。定心胶囊治疗组、维拉帕米对照组给药治疗,4周后,测定血清中P-选择素的含量。结果:①治疗组(包括西药、定心胶囊组)与模型组比较,治疗组血清中P-选择素含量明显降低,P<0.01,说明有显著意义,治疗有效。②定心胶囊组与西药对照组比较,P>0.05,没有统计学意义,说明定心胶囊与西药维拉帕米对房颤的治疗与预防作用是相近的。结论:定心胶囊能够降低血清中P-选择素的含量,减低血小板活化,对房颤大鼠血栓形成和血栓栓塞有预防和治疗作用。Objective： The aim of this study is to develop the effective therapy for atrial fibrillation （AF）, and study the mechanisms of Dingxin capsule. To elucidate the role of platelet P-selection in the pathogenesis of AF. Methods ： 36 rats were randomized to 4 groups ： normal control group, model group, Dingxin capsule cu^atio group, Verpamil intervention group. Plasma P-selection was measured with ELISA. Results： With the comparison of treating group and model group, the level of P-selection in treating group lowed obviously （ P 〈0.01 ）. It had statistic significance. The express level of platelet P-selection of Dingxin capsule curatio group and the Verpamil intervention group have similar effect to AF （ P 〉0.05）. It had not statistic significance. Conclusion： The research results showed that Dingxin capsule could low the expression of P-sdection, which played important function in the treatment of AF.
