尿酸利仙含药血清对尿酸盐诱导的人血管内皮细胞VCAM-1 mRNA表达的影响

摘　　要：目的:观察中药尿酸利仙(含药血清)对尿酸盐诱导的人血管内皮细胞(Endothelial cell of vessels,ECV)血管细胞黏附分子-1(VCAM-1)mRNA表达的影响。方法:①体外培养人血管内皮细胞,用终浓度为1070μmol/L的尿酸盐溶液刺激,同时用低、中、高浓度(2.5%、5%、10%)尿酸利仙含药血清的培养液进行干预,用逆转录-聚合酶链式反应(RT-PCR)技术测定细胞VCAM-1的mRNA表达。结果:尿酸盐可诱导血管内皮细胞VCAM-1的mRNA高表达,差异有极显著的统计学意义(P<0.01);各浓度尿酸利仙含药血清干预组细胞VCAM-1的mRNA表达均较尿酸盐刺激组下降,差异有统计学意义。结论:尿酸利仙含药血清可抑制VCAM-1过度表达,是减轻尿酸盐诱导的人血管内皮细胞炎症损伤及抑制炎症反应的机制之一。Objective：To investigate the effect of the herbal compound （NiaoSuanLiXian） on the expression of Vascular cell ad- hesion molecule （VCAM-1） in vascular endothelial ceils. Methods ：Cultured the vascular endothelial ceils, and stimulated with urate（ 1070p, mol/L）. At the same time, the ceils were co-incubated with the serum including medicine. The expression of VCAM-1 in the vascular endothelial ceils was detected by FCM and RT-PCR. Results ：The expression of VCAM-1 in the vascular endothelial cells induced by urate increased significantly than that in the control cells. 2.5% ,5% , 10% of the serum including NiaoSuanLiX- ian could all significantly down-regulate the expression of VCAM-1 on levels of mRNA. Conclusion： Down-regulating the expression of VCAM-1 in vascular endothelial ceils induced by urate is the One of mechanism of NiaoSuanLiXian reducing the inflammatory injure.
