摘 要:目的 研究慢性应激抑郁模型大鼠海马中NPY的表达情况及氟西汀的干预作用。方法将大鼠随机分为抑郁模型组、氟西汀治疗组和对照组。模型组和氟西汀组给予21d的应激刺激,此期间对照组正常饲养,刺激期间氟西汀组每天腹腔注射氟西汀(10mg/kg),模型组和对照组每天腹腔注射等体积的生理盐水。行为学检测应用open-field法和液体消耗实验。采用western-blotting法检测各组大鼠海马NPY表达的水平。结果 应激后模型组水平穿越格数(19.5士7.2)、竖立次数(10.2±2.6)、修饰次数(6.6±2.4)、糖水消耗百分比[(53.1±9.5)%],均显著低于对照组[分别为(31.4±7.3)、(19.0±2.4)、(14.0士2.4)、(76.0±2.7);均P〈0.01]。应激后氟西汀组水平穿越格数(26.2±3.5)与对照组差异无显著性(P〉0.05),但高于模型组(P〈0.05);竖立次数(13.9±4.2)低于对照组(P〈0.01),但高于模型组(P〈0.05);修饰次数(10.3±1.6)低于对照组(P〈0.01),但高于模型组(P〈0.01);糖水消耗百分比(61.5±8.5)%低于对照组(P〈0.01),与模型组差异无显著性(P〉0.05)。模型组大鼠慢性应激后海马中NPY的表达水平显著低于对照组(P〈0.01));氟西汀组大鼠慢性应激后海马中NPY的表达水平显著低于对照组(P〈0.01)),但明显高于模型组大鼠,差异有显著性(P〈0.01)。结论 慢性应激抑郁模型大鼠海马中NPY的表达降低并可以被氟西汀部分逆转。Objective To research the expression of NPY in hippocampus of chronic stress depression rat and the interference of fluoxetine to the NPY. Methods All the experimental rats were divided by random into 3 groups : Group of depression, Group of fluoxetine and Group of control. The rats of Group of depression and Group of fluoxetine were applied stress for 21 days, and meanwhile the rats of Group of control were fed normally. The rats of Group of fluoxetine were applied fluoxetine by peritoneal injection ( 10mg/kg), and the rats of another groups were given normal sodium of the same volume by peritoneal injection. The ethology examination was performed by using method of open-field and experiment of fluid consumption. The expression of NPY was detected by Western-blotting. Results After the chronic stress, the horizontal crossing numbers, the erection times, the mod- ification times and the percentage of sacchar-consumption of the rats of Group of depression are 19.5 ± 7.2;10.2 ± 2.6 ;6.6 ± 2.4 and 53.1% ± 9.5% respectively, which are less than Group of control ( 31.4 ± 7.3 ; 19.0 ± 2.4 ; 14.0 ± 2.4 and 76.0 ± 2.7 ; P 〈 0.01 ). There is no obvious difference of horizontal crossing numbers between Group of fluoxetine and Group of control( P 〉 0.05), but more than Group of depression ( P 〈 0.05 ). As far as the erection times concerned, Group of fluoxetine is less than Group of control ( P〈0.01 ) while more than Group of depression ( P 〈 0.05) ; the modification times, Group of fluoxetine is less than Group of control ( P 〈 0.01 ), while more than Group of depression ( P〈0.01 ) ; the percentage of sacchar-consumption, Group of fluoxetine is less than Group of control ( P〈0.01 ), however compared with Group of depression, the difference is not significant ( P 〉 0.05 ).
