摘 要:目的:本实验研究是为了探讨中药复方制剂增骨丸对维甲酸诱导的大鼠骨质疏松模型骨组织显微结构的影响,为增骨丸治疗原发性骨质疏松症提供实验依据,为今后骨组织形态计量学在此领域进一步发展提供资料。方法:50只8月龄雌性SD大鼠,随机分为2组:正常对照组(13只),模型组(37只)。模型复制成功后,把模型组随机分为三组:骨质疏松组、乙烯雌酚组、增骨丸组。治疗两个月,观察增骨丸对骨质疏松模型大鼠骨组织形态计量学的影响。结果:增骨丸组、乙烯雌酚组与骨质疏松组比较:静力学参数BV/TV、Tb.Th、Joint均显著增加,而FLAW明显减少,OBI明显上升;动力学参数dLs/BS、BFR也显著增加,动力学参数MLT缩短。但是静力学参数BV/TV、Tb.Th、OV/BV增骨丸组比乙烯雌酚组明显升高,动力学参数MAR、BFR增骨丸组比乙烯雌酚组明显下降。结论:增骨丸类激素样作用,可增加成骨细胞的数量和活性,明显提高成骨作用,类骨质矿化的速度加快,使骨量增加,改善骨组织形态计量学作用,可改善骨质疏松的有效药物。Objective:To investigate the effect of Zenggu pills(zgp) on histomorphometry in osteoporotic rats models induced by retinoic acid in order to provide experimental basis for clinical application of the drug and further study. Methods: Fifty healthy female SD rats of eight months old were randomly divided into the normal group and osteoporotic group. The model was successfully established by retinoic acid. Osteoporotic group were divided into the model group,diethylstilbestrol(DES) treated group and ZGP treated group, The rats were hyped with muriatic tetracycline liquor to make double fluorescence sign on labelling the 13rd and 3rd day before being killed untill two months . The tibias was then picked out and fixed ,embeded,and cut into sections in order to have an image analys. Results:Compared osteoporotic group,ZGP treated group and diethylstilbestrol(DES) treated group: static parameter BV/TV, Tb. Th, Joint , OBI obviously increased, FLAW distinctly failed;the dynamic parameter dLs/BS and BFR obviously increased, MLT obviously shortened. Compared with diethylstilbestrol(DES) treated group, ZGP treated group static parameter BV/TV, Tb. Th, OV/BV obviously increased ; dynamic parameter MAR and BFR obviously failed. Conclusion:ZGP has the therapeutic effects on osteoporosis . It's mechanisms is to increase the E2 level , preventing the bone resorption and influencing the bone formation. There is a good prospect for clinical application.
