摘 要:目的探讨经颅低频脉冲磁场对全脑缺血再灌注大鼠海马神经细胞的作用。方法将24只SD大鼠随机分成对照组和磁辐射组,每组各12只,应用改良的Pulsineli4-VO法制做大鼠全脑缺血再灌注模型,磁辐射组在脑缺血模型完成后立即将其头颈部置磁场(20mT、10Hz)中轴中央,身体顺磁场方向放置,辐射45min,每天1次,作用4次后将大鼠记录脑电图后断头取脑,对照组同时处死断头取脑,应用Nissl染色及免疫组织化学方法进行海马神经细胞计数及凋亡相关基因caspase-3蛋白表达检测。结果磁疗组较对照组神经元数量多(P〈0.05),脑电图频率和波幅增高(P〈0.05),caspase-3蛋白表达低(P〈0.05)。结论低频脉冲磁场(20mT、10Hz)可以抑制大鼠脑缺血再灌注海马神经元的凋亡,对脑缺血再灌注损伤有保护作用。Objective To investigate the effects of low-frequency pulse electromagnetic field (LFPMF) on hippocampal neurons following the global cerebral ischemia/reperfusion (I/R) in rats. Methods Twenty-four adult male Sprague-Dawley rats were randomly divided into control and radiation groups (n=12 respectively). The global I/R model was established by the modified Pulsineli 4 vessel occlusion (4-VO) method. After modeling, the rats in radiation group were exposed to LFPMF (20 mT, 10 Hz) for 45 min per day for 4 d, and all the rats were sacrificed at 4th day. The electroencephalogram (EEG) was recorded by Powedab system; Nissl staining was used for neuron counting and immunohistochemical staining was employed to detect the expression of apoptosis-associated caspase-3 gene. Results The number of neurons in the hippocampal CA1 area in the radiation group was significantly higher than that in the control group (P〈0.05). The amplitude and frequency of the EEG were higher in the radiation group than those in the control group (P〈0.05). The positive rate of caspase-3 in the hippocampal CA1 area in the radiation group was lower than that in the control group (P〈0.05). Conclusion The LFPMF (20 mT, 10 Hz) treatment has neuroprotective effect on the rats with global cerebral I/R injury through restraining the neuronal apoptosis involved in global cerebral I/R.
