摘 要:目的探讨脑内c-fos基因表达及血浆β-内啡肽的变化在二次脑损伤(SBI)中的作用。方法125只雄性SD大鼠随机分为正常对照组、假手术组、脑缺血组、颅脑损伤组和SBI组。分别采用免疫组化及放免分析测定大鼠脑内c-fos蛋白与血浆β-EP的含量,并对脑组织进行病理学观察。结果SBI组c-fos基因表达分别于伤后3h、24h出现两个高峰,伤后3 ̄48hc-fos基因表达均明显高于颅脑损伤组及脑缺血组(P<0.05)。SBI组大鼠血浆β-EP水平在伤后1h达高峰,至伤后48h仍未恢复正常(P<0.05),伤后各时间点均明显高于颅脑损伤组和脑缺血组(P<0.05)。在伤后12hSBI组脑含水量达高峰;伤后3 ̄48h明显高于颅脑损伤组及脑缺血组(P<0.05)。伤后6 ̄48hSBI组神经元损伤数目明显高于颅脑损伤组及脑缺血组(P<0.05)。结论伤后大鼠脑内c-fos基因表达增加、血浆β-EP水平明显升高参与了SBI后病理生理过程,提示两者同SBI发展密切相关。Objectives To explore c-fos gene expression and change in plasma level of β-endorphin (β-EP) after secondary brain insults(SB1) and their significance in rats. Methods One hundred and twenty-five male SD rats were randomly divided into five groups, i.e. normal control, sham-operation, cerebral ischemia, traumatic brain injury (TBI) and SBI groups. The expression of the c-fos gene in the brain tissues was detected by immunohistochemistry and the plasma level of beta-endorphin was determined by radioimmunoassay. The determination of water contents and the histopathological examination of the brain tissues were performed. Results The two peaks of c-fos gene expression in the brain tissues appeared 3 and 24 hours respectively after the injury. The rate of e-fos positive cells in SBI group was significant higher than those in cerebral ischemia and TBI groups from 3 to 48 hours after injury (P〈0.05). The plasma level of β-EP in SBI group reached the peak 1 hour after the injury and were significantly higher than that in cerebral ischemia and TBI groups from 1 to 48 hours after the injury (P〈0.05). The water contents in the brain tissues in SBI group reached the peak 12 hours after the injury and were significantly higher than those in cerebral ischemia and TBI groups from 3 to 48 hours after injury (P〈0.05). The number of damaged neurons of the brain tissues in SBI group significant was more than those in cerebral ischemia and TBI group from 6 to 48 hours after the injury (P〈0.05). Conclusion The increases in c-fos gene expression and plasma level of β-EP may take part in the pathophyslological process and development after the SBI.
