摘  要:采用丙型肝炎病毒(HCV)5’-非编码区(5’-NCR)和C区具有型特异性的引物建立的逆转录-聚合酶链反应方法检测了25例丙型肝炎(HC)和26例抗-HCV阳性献血员的HCV RNA及基因型。结果25例HC患者HCV RNA阳性率为100.0%,26例抗-HCV阳性献血员HCV RNA阳性率为84.5%,(22/26)。25例HC中Ⅰ、Ⅱ和混合型(Ⅰ+Ⅱ、Ⅱ+Ⅲ、Ⅰ+Ⅲ、Ⅰ+Ⅱ+Ⅲ)分别占8.0%、16.0%、和64.0%。其中Ⅰ+Ⅱ占44.0%,未定型3例占12.0%。22例HCV RNA阳性献血员中Ⅰ型占13.6%(3/22),Ⅱ型占59.1%(13/22),混合型(Ⅰ+Ⅱ、Ⅱ+Ⅲ、Ⅰ+Ⅲ、Ⅰ+Ⅱ+Ⅲ)各1例占18.0%(4/22),未定型2例。二组均未发现Ⅳ型。In order to investigation the relationship of virus genotypes between post-transfused hepatitis C (HC) and anti-HCV positive blood donors, the HCV RNA and genotypes were detected in 25 HC cases and 26 anti-HCV positive blood donors by RT-PCR with a pair of primers according to the sequence of HCV 5'-Noneoding Region (5'-NCR) and primers for genotyping according to the sequence of C region. The results were as follows: HCV RNA positive rate in all 25 (HC) cases was 100% and in 26 positive blood donors was 84.5% (22/26). The HCV genotypes in 25 HC cases were as follows: 2 cases were type Ⅰ, 4 cases were type Ⅱ; 16 case were coinfected among them 11 cases were infected by type Ⅰ + Ⅱ, two case by Ⅱ+ Ⅲ and Ⅰ+Ⅱ+Ⅲ respectively one case by Ⅰ+Ⅲand 3 cases remained undetermined. HCV geno-types of 22 blood donors were: 3 cases for type Ⅰ, 13 cases for type Ⅱ, 4 cases were coinfectied (one case for type Ⅰ+Ⅱ, type Ⅰ+Ⅱ+Ⅲ, type Ⅰ+Ⅲ, and type Ⅱ+Ⅲ each) and 2 cases were undetermined. HCV genotype Ⅳ was not detected in the two groups. Comparison of distribution of HCV genotypes in two groups showed that HCV type Ⅱ was the most common genotype in the blood donors whereas coinfection of type Ⅰ+Ⅱ were predominant in HC patients. This paper indicated that HCV RNA positive blood donors are the main infection source of hepatitis C virus, the more transfusion, the more high genotypes coinfection. Serum samples blood donors of should be strictly screened to test anti-HCV to prevent the HC.
