快速免疫诊断牛布鲁氏菌病新方法的研究

摘　　要：目的建立一种牛布鲁氏菌病快速免疫诊断方法。方法斑点金免疫渗滤法（DIGFA）以硝酸纤维素膜为载体，胶体金标记蛋白为显示剂，抗原、抗体及胶体金标记蛋白通过渗滤发生反应，阳性结果在膜上显示红色斑点。结果DIGFA快速检测牛布鲁氏菌病具有很好的重现性和稳定性；敏感性、特异性分别达到97％和100%。结论DIGFA快速检测牛布鲁氏菌病，方法快速简便，测试过程1～2min内完成，试剂敏感、特异、稳定，非常适合于布鲁氏菌病的诊断及流行病学调查。The dot immuno-gold filtration assay （DIGFA） was used in the present study, in which nitrocellulose membrane was used as carrier, protein labeled with colloidal gold used as probe , and antigen, antibody and protein labeled with colloidal gold combined together through filtration with appearance of red dots on the membrane as a positive reaction. This method had been applied for the detection brucella in cows with excellent reproducibility and stability. Its sensitivity and specificity were 97% and 100% respectively and course of performance was quite simple and rapid, completing the whole course of testing within 1-2 minutes. Therefore, it is suitable to the clinical diagnosis and sero-epidermiological survey for cows with brucellosis.
