清金得生片对人肺腺癌细胞(LAC)抑制与凋亡的诱导作用

摘　　要：【目的】观察清金得生片对人肺腺癌细胞(LAC)的抑制与凋亡的影响。【方法】以8 g/kg剂量的清金得生片,0.2 g/kg剂量的替加氟(FT-207)灌胃SD大鼠制备体积分数为10%、20%、30%的中药含药血清和体积分数为20%的FT-207含药血清,并设空白血清对照;采用四甲基偶氮唑盐(MTT)染色法观察清金得生片对LAC的抑制作用;采用硫氰酸荧光素(FITC)/碘化丙啶(PI)双染色法,以流式细胞仪检测清金得生片对LAC细胞早期凋亡的影响;采用透射电镜观察中药对LAC超微结构的影响。【结果】不同剂量中药含药血清对LAC增殖的抑制作用与药物浓度及作用时间呈一定依赖关系,其中体积分数为30%的中药含药血清作用72 h时的抑瘤率达32%;各剂量组中药含药血清均具有一定诱导LAC细胞凋亡的作用;中药含药血清高、中剂量组透射电镜下可见明显凋亡特征,低剂量组凋亡特征不明显。【结论】清金得生片的抗肿瘤机制可能与其能抑制LAC增殖、诱导细胞凋亡作用有关。[Objective] To observe the effect of Qingjin Desheng Tablets （QDT） on inhibiting proliferation and inducing apoptosis of human lung adenocarcinoma cells （LAC）. [ Methods ] Serum containing QDT at different concentrations （10%, 20% and 30% respectively） was obtained from SD rats which received gastric gavage of 8 g/kg QDT. Serum containing Tegafur （FT-207） at the concentration of 20% was obtained from SD rats which received gastric gavage of 0.2 g/kg FT-207. Meanwhile, the blank control serum was also prepared. The inhibitory effect of QDT on LAC proliferation was observed with methylthiazolyltetrazolium （MTT） assay. Annexin V-fluorescein isothiocyanate/propidium iodide （Annexin V-FITC/PI） double-labeled method and flow cytometry （FCM） were used to detect the early apoptosis of LAC, and the changes of LAC ultrastructure was examined under transmission electron microscope. [ Results] Serum containing QDT inhibited LAC proliferation in a dose-time dependent manner, and the tumor-inhibitory rate of serum containing QDT at the concentration of 30% arrived 32% after 72-hour culture. Serum containing QDT also had an effect on inducing LAC apoptosis. And apoptotic features were obvious under transmission electron microscope in high and moderate-dose groups, while not obvious in low-dose group. [ Conclusion] The anti-tumor mechanism of QDT may be related to the inhibition of LAC proliferation and to the induction of LAC apoptosis.
