摘  要:目的探讨大鼠移植心脏心肌细胞凋亡的机制及ZnSO4在心脏移植早期抗心肌细胞凋亡的效果。方法将Ono大鼠腹腔心脏移植模型共18例分为3组,A组:术后受体每隔12 h腹腔注射生理盐水1 m l;B组:术后受体每隔12 h按2 mg.kg-1腹腔注射0.31 mmol.L-1ZnSO4溶液;C组:术后受体每隔12 h按5 mg.kg-1腹腔注射0.31 mmol.L-1ZnSO4溶液。术后第4天取移植心脏右心室心肌组织进行细胞凋亡检测和caspase-3免疫组化染色。结果各组移植心脏均可见细胞凋亡的存在,各组凋亡指数组间均存在显著差异(P<0.05)。各组均可见caspase-3表达阳性的细胞存在,各组caspase-3表达指数组间比较存在显著差异(P<0.05)。结论caspase-3与心脏移植术后的心肌细胞凋亡之间关系密切。ZnSO4通过影响caspase-3的活化而发挥抗移植心脏心肌细胞凋亡效应。Objective To observe the apoptotic mechanism and anti-apoptosis effects of ZnSO4 in rat heart transplantation model at early stage. Methods A rat heterotopic cardiac transplant model with Wistar recipients and SD donors was constructed , Group A was treated with 1 ml physiological saline injection peritoneum eyery 12 h after heart transplantation ;group B was performed with 0.31 mmol ·L^-1 ZnSO4 (2 mg ·kg^-1 ) injection peritoneum every 12 h after heart transplantation, group C was conducted with 0.31 mmol ·L^-1 ZnSO4 (5 mg ·kg^-1 ) injection peritoneum every 12 h after heart transplantation and at the 4th day after operation, immunohistochemical and terminal deoxynucleotidyl transferse-mediatedte (TUNEL) techniques were used to detect myocardial apoptosis and caspase-3-positive cell. Results It demonstrated apoptosis in every group. There were significant difference in apoptotic indexes among the three groups( P 〈 0.05 ). There were demonstration of caspase-3 positive cells and there were significant difference of caspase-3 expression among the three groups. Conclusion The activation of caspase-3 is correlated with cell apoptosis in heart transplantation. ZnSO4 plays an important role in antiapoptosis of cardiac muscle cells by impacting activation of caspase-3.
