治喘贴对大鼠免疫球蛋白E和嗜酸粒细胞凋亡的调节作用

摘　　要：目的探讨治喘贴对卵清蛋白所致大鼠IgE及嗜酸粒细胞(EOS)凋亡的调节作用。方法将大鼠随机分为正常对照组、哮喘模型组、代温灸膏治疗组、治喘贴治疗组。经外贴治疗18d,收集支气管肺泡灌洗液(BALF)并采集心脏血,采用免疫组织化学(TUNEL)和酶联免疫吸附(ELISA)法分析血中IgE含量和观察EOS凋亡及BALF中EOS凋亡数。结果哮喘模型组血和BALF中IgE含量和EOS凋亡数均高于正常对照组动物,治喘贴治疗组大鼠IgE和EOS均明显低于哮喘模型组;光镜下观察,治喘贴治疗组EOS凋亡数目较多,肺炎症程度较轻,支气管壁形态较稳定。结论治喘贴能降低IgE水平,并促进哮喘动物体内EOS凋亡,从而改善哮喘模型实验动物的气道变态性炎症反应。Objective： To investigate the effect of Zhichuan Plaster（治喘贴） on the levels of IgE and eosinophils （EOS） apoptosis in rats infected with ovalbumin. Methods： Rats were randomly divided into the fitness group, the asthma group, the Daivenjiu Plaster（代温灸膏 ） treatment group and the Zhichuan Plaster treatment group. After 18 days＇ treatment, collected the bronchus alveoli lavage fluid（BALF）and the heat blood of rats. Then assayed the level of IgE, the apoptosis of EOS, and the quantity of apoptosis EOS in BALF through terminal deoxynuclectidyl transferase - mediated dUTP nickend - labeling （TUNEL） and enzyme - linked immunosorbent assay（ELASA） . Results： The levels of IgE, apoptosis EOS in blood and BALF in the asthma group were higher than which in the fitness group. Those of the Zhichuan Plaster treatment group was lower than of the asthma group. Conclusions： Zhichuan Plaster can reduce the levels of IgE, EOS, and it has an apoptosis effect on EOS. It can improve ainvay allergy inflammation of asthma in rats with avalbumin.
