摘  要:目的:观察咳喘宁对支气管哮喘大鼠肺组织一氧化氮(NO)和内皮素(ET-1)含量的影响。方法:40只SD大鼠随机分为正常组、模型组、咳喘宁高剂量组(27g生药/kg体质量)、咳喘宁低剂量组(13.5g生药/kg体质量)、桂龙咳喘宁组(0.41g/kg体质量),每组8只。除正常组外以卵蛋白致敏并吸入激发法制备大鼠支气管哮喘模型,各治疗组均从第1次哮喘激发开始(造模第3周)至处死前每天灌胃给药,激发并给药4周后处死大鼠,观察肺组织NO、ET-1含量和病理组织学变化。结果:与正常组比较,模型组大鼠支气管管壁和平滑肌厚度、嗜酸性粒细胞、淋巴细胞数以及肺组织NO、ET-1含量明显增加(P<0.01);与模型组比较,各治疗组均可显著降低支气管管壁和平滑肌厚度、嗜酸性粒细胞、淋巴细胞数以及肺组织NO、ET-1含量(P<0.05或P<0.01);且咳喘宁高、低剂量组优于桂龙咳喘宁胶囊组(P<0.05或P<0.01)。结论:咳喘宁可通过降低肺组织NO和ET-1含量,抑制支气管哮喘大鼠气道炎症和气道重塑等病理过程。Objective: To observe the effects of Kechuanning on nitrogen monoxidum (NO) and endothelin (ET-1) of in pulmonary tissue of rat with bronchial asthma. Methods: 40 rats were randomly divided into 5 groups: the normal group, the model group, Kechuanning high-dose group (27g/kg), Kechuanning low-dose group (13.5g/kg) and Guilong Kechuanning Capsule control group (0.41g/kg), 8 rats each group. The bronchial asthmatic model was established by egg protein sensibilization and long-term inhalation provocation. The rats of each treatment groups were given intragastric administration each day from the first time of provocation to execution. After 4 weeks of treatment,the rats were killed and lung tissue were taken to dyeing of HE to be observed. The contents of NO and ET-1 in lung tissue were determined. Results: Compared with the normal group, the thickness of bronchus wall and bronchus smooth muscle, the numbers of eosinophile granulocyte and leukomonocyte, and the contents of NO and El'- 1 in lung tissue were increased in the model group ( P 〈 0.01) ; Com- pared with the model group, the thickness of bronchus wall and bronchus smooth muscle, the numbers of eosinophile granu- locyte and leukomonocyte and the contents of NO and ET-1 in lung tissue were decreased in the each treatment group ( P 〈 0.05 or P 〈 0.01); And Kechuanning high-dose and low-dose groups surpass Guilong Kechuanning Capsule group( P 〈 0.05 or P 〈 0.01). Conclusion: Kechuanning could decrease the contents of NO and ET- 1 in lung tissue, so as to restrain air passage inflammation and against airway remolding.
