摘  要:多花黑麦草斑驳病毒(Ryeg rass mottle virus,RGM oV)属南方菜豆花叶病毒属,是牧草病毒病的重要病原。利用反转录技术合成和扩增了RGM oV RNA的cDNA。将cDNA克隆在载体pUC18上,对RGM oV的基因组RNA作序列分析。RGM oV RNA是一个正义单链RNA分子,全长4 210个核苷酸,包含4个开放阅读框架5′非翻译区包含99个核苷酸,3′非翻译区有198个核苷酸。RGM oV ORF的结构和南方菜豆花叶病毒以及水稻黄斑病毒的结构一样。ORF1编码1个14.6 kD的蛋白质,这个蛋白质的功能还不清楚;ORF2的产物由947个氨基酸组成,分子量为103.6 kD;ORF3编码1个19.8 kD的蛋白质;ORF4编码1个25.6 kD的蛋白质,通过N末端氨基酸分析,确认ORF4为病毒外壳蛋白。RGM oV RNA的全序列以及染色体结构已被GenBank登录,登录号分别为AB040446和NC_003747。As a member of Sobemovirus, Ryegrass mottle virus (RGMoV) is an important herbage grass pathogen. The cDNA fragment was synthesized and amplified by reverse transcription, and cloned into pUC18 for sequencing. The genome of RGMoV was a positive-sense ssRNA of 4 210 nucleotides in length as revealed by sequencing the entire genome. Its coding region contained four open reading frames (ORFs), the 5'-untranslated region of the genome with 99 nucleotides and the 3'-untranslated region with 198 nucleotides. The structure of RGMoV ORFs was similar with that of the previously sequenced Southern bean mosaic virus and Rice yellow mottle virus. ORF1 encoded a protein that had a calculated molecular mass of 14.6 kD and the function of this protein was unknown. The ORF2 product consisted of 947 amino acids, with a calculated molecular mass of 103.6 kD. ORF3 encoded a protein having a calculated molecular mass of 19.8 kD. ORF4 encoded a 25.6 kD coat protein which was verified by amino acid sequencing of the RGMoV coat protein N terminus. The nucleotide sequence and genome structure datum reported in this paper had been deposited with EMBL databese under accession number AB040446 and NC_003747.
