肠露灌肠剂治疗小鼠免疫性溃疡性结肠炎的实验研究

摘　　要：目的观察中药复方肠露灌肠剂对小鼠免疫性溃疡性结肠炎(ulcerative colitis,UC)模型的治疗作用。方法将实验小鼠随机分为正常组、模型组、肠露高剂量组(高剂组)、肠露低剂量组(低剂组)、柳氮磺吡啶(SASP)组。除正常组外,其余小鼠用兔结肠黏膜蛋白免疫法制作实验UC模型。给药治疗21天后,观察各组小鼠的疾病活动指数、体重变化,然后处死小鼠,取全段结肠,称结肠湿重,测肠重指数,比较各组小鼠的疾病活动指数、体重变化、结肠炎症反应及结肠组织病理学改变。结果肠露高、低剂量组,SASP组疾病活动指数明显减少,体重改善,结肠炎症反应减轻,与模型组比较差异有显著性(P<0．05或P<0．01);病理检查示:模型组小鼠结肠黏膜糜烂及溃疡形成、炎细胞浸润明显,肠壁充血伴出血,水肿明显;SASP组小鼠结肠黏膜未见明显糜烂及溃疡形成,但炎细胞浸润较明显,肠壁轻度充血,轻度水肿;肠露高刺组小鼠结肠黏膜无糜烂及溃疡形成,部分炎细胞浸润,肠壁轻度充血、无水肿。结论中药复方肠露灌肠剂对小鼠免疫性UC有明显的疗效。Objective To observe the therapeutic effect of Chinese compound, Changlu Enema （CE）, on immune ulcerative eolitis （UC） in miee. Methods Mice were randomly divided into the normal group, the model group, the CE high dose （CE-H） and low dose （CE-L） group and the salicylazosulfapyridine （SASP） group. All mice, exeept those in the normal group, were made into UC model by eolonic mueosa protein immunization. After 21 days of medieation, the changes of UC activity index and body weight in mice were observed, and the condition of eolonic inflammation and histomorphologieal changes in eolonie tissue were observed also. Results UC activity index was lower and body weight was higher in the two CE groups than those in the model group respectively, showing significant difference （P 〈 0.05 or P 〈 0.01） ; pathological examination showed the pathological changes and inflammatory response in colonic tissue were relieved significantly after treatment, and the improvement in the CE-H group was better than that in the SASP group. Conclusion TCM compound CE is markedly effective in treating UC rats.
