摘  要:根据大鼠细胞因子信号转导抑制因子(suppressorofcytokinesignaling,SOCS)-3设计并合成一对引物,从八眉猪皮下脂肪组织提取总mRNA,RT-PCR扩增获得猪SOCS-3cDNA;利用半定量(semi-quantitative,SQ)RT-PCR技术检测大白猪各组织中SOCS-3mRNA的表达量.扩增获得SOCS-3基因GenBank登陆号DQ644577,用CDD软件进行SOCS-3基因结构分析,该序列具有SOCS-3特有的保守结构域SH2和SOCS-box.用ClustalW软件进行同源性分析发现,猪SOCS-3基因与人、大鼠和小鼠的同源性分别为92%、88%和88%.SQRT-PCR组织表达检测表明SOCS-3基因在心脏、肝脏、脾脏、肺脏、肾脏、肌肉、皮下脂肪和内脏脂肪中均有表达,其中肺脏和脾脏的表达丰度最高,肝脏和内脏脂肪中的表达量最低.A pair of PCR primers were designed and synthesized according to rat' s SOCS- 3 gene sequence. Total RNA was isolated from adipocytes of Bamei pig and reverse-transcripted to cDNA, which was then subjected to SQ (semi-quantitative) RT-PCR. RT-PCR product was sequenced and submitted to GenBank (access number DQ644577). The SOCS-3 cDNA sequence was analyzed by Conserved Domain Database (CDD) and ClustalW software respectively. The results showed that the sequence contained two conservative regions(SH2 and SOCC-box) and had 92% homology to that of human,88% homology to that of rat and 88% to that of mouse. Also, we found that SOCS-3 gene was widely expressed in the heart, liver, spleen, lung, kidney, muscle, subcutaneous adipose and visceral adipose tissues. Its expression level was relatively higher in lung and spleen and lower in live and visceral adipose.
