High Performance Liquid Chromatographic Determination of Dissolved Free Amino Acids in Seawater

摘  要:采用邻苯二甲醛柱前衍生-高效液相色谱-荧光检测器(OPA—HPLC-FLD)联用技术分离测定了海水中的14种主要溶解游离氨基酸(DFAA),通过邻苯二甲醛-3-巯基丙酸(OPA-3-MPA)柱前衍生后,在XDB—C18柱上分离。不同氨基酸测定的精密度(RSD)为1.62%~3.80%,检测限为11.4~69.7ng·L^-1,海水加标回收率在92%~110%之间。利用该方法测得青岛近海表层海水中氨基酸浓度为0.015~0.284μmolL^-1。A system using reversed-phase high-performance liquid-chromatography coupled with fluorescence detection(RP-HPLC-FLD)has been developed for the determination of 14 main dissolved free amino acids in seawater. The free amino acids were derivatized with O-Phthalaldehyde and 3-mercaptopropionic acid (OPA-3- MPA). The resulting OPA derivatives were then chromatographed on an XDB-C18 column. The measurement precisions for different amino acids were between 1.62 and 3.80% (relative standard deviation); the detection limits were between 11.4 and 69.7 ng L^-1 ; the recoveries of standards added to seawater samples were between 92 and 110%. The amino acid concentrations in Qingdao coastal seawater determined with this system ranged( from 0. 015-0. 284 μmol L^-10.
