摘  要:目的建立犬心肌梗死动物模型并加以评价。方法选取杂种犬14只,建立模型前进行心电图、超声及SPECT检查;常规麻醉实验犬,开胸结扎左冠状动脉前降支,同时作心电监护;11 d后行超声心动图及SPECT检查。结果犬冠状动脉前降支结扎后,心电监护示心率加快和室性早搏,ST段压低;11 d后超声心动图显示有室壁活动减弱,心梗区心肌变薄,射血分数(EF)、每搏量(SV)、左室短轴缩短率(FS)下降,较结扎前差异有统计学意义(P<0.05);99rnTc-MIBI示:心尖部、左室前壁、室间隔明显充盈缺损。结论该方法效果确实可靠、手术方便安全,是构建心肌梗死模型的理想方法。Objective To develop and evaluate the canine model of myocardial infarction.Methods Fourteen mongrel dogs were used.At first,electrocardiography (ECG), ultrasoundcardiography(UCG) and ^99mTc - MIBI positron emission tomography(SPET) were performed to obtain the baseline data. Canines were anesthetized with penthiobarbital sodium and heart was exposed via a left lateral thoracotomy, acute inyocovrdial infarctim (AMI) was generated by ligating the left anterior descending coronary artery. At the same time, the dogs were examined by ECG. Eleven days later, UCG and SPEF were examined to confinn the development of AMI canine model. Results After the left anterior descending coronary was ligated, the electrocardiogram showed tachycardia, ST- segments depressed in limb leads, and ventricular premature beat. Eleven days after the development of AMI, UCG demonstrated dyskinesis at anterior wall segment d the heart. The left ventricuhr ejection fraction(EF), stroke vohme(SV), and fractional shortening(FS) were lower than those preoperafion( all P 〈 0.05). ^99mTc - MIBI myocardial perfusion imaging showed that fixed perfusion defect appeared in apical, anterior and septal wall of model canines. Conclusions The procedure can easily develop the myocardial infarction model dog with effective results, and no expensive instruments are needed.
