孕酮和A23187对精子顶体反应、活力和活率的影响

摘　　要：目的研究精子顶体反应诱导剂孕酮和A23187对顶体反应(Acrosomereaction,AR)、精子活力和活率的影响。方法10例健康生育男子精液,采用Percoll密度梯度分离法优选精子,用FITC-PSA荧光染色法分析AR,用伊红Y染色法分析精子活率,用精子自动分析仪分析精子活力。结果当A23187终浓度为5、10、20及40μmol/L时,顶体反应率均明显增加,与对照组比较,差异均有显著性(P<0.05)。随着A23187浓度增加,精子活力和活率均明显下降(P<0.01)。当孕酮终浓度为5、10、20及40μg/mL时,顶体反应率均明显增加,与对照组比较,差异亦均有显著性(P<0.05)。但随着孕酮浓度增加,精子活力和活率与对照组比较差异均无显著性(P>0.05),与A23187组比较,差异均有显著性(P<0.001)。结论孕酮诱导顶体反应过程不影响精子活力和活率,与A23187比较更接近生理过程。[Objective] The effects of progesterone and calcium ionosphere A23187 on AR and sperm motility, viability were studied. [Methods] Human semen was obtained from 10 feaile donors with normal semen parameters. Specimens were allowed to liquefy at 37%, and were prepared by Percoll-gradient treatment. Spermatozoa were recovered from the upper layer. Acrosome reaction was analyzed using FITC-PSA staining. The sperm viability was analyzed using eosin staining. [Results] When the final concentration of calcium ionosphere A23187 were 5, 10, 20 and 40 μm/L, the percentage of acroseme reacted sperm were significantly （P 〈0.05） increased. With increase of concentration of A23187, the percentage of sperm motility and viability markedly decreased （P〈0.01）. When the final concentration of progesterone were 5, 10, 20 and 40μg/mL, the percentage of acrosome reacted sperm was significantly （P 〈0.05） increased. The percentage of sperm motility and viability were not different in comparison with antitheses group （P 〉0.05）, but there were markedly differences in comparison with A23187 group （P 〈0.001）. [Conclusion] Progesterone-induced AR doesn＇t influence sperm motility and viability. It is even more approximate physiological process.
