Effects of Oenotheria binnis seed oil on lipoperoxides, plasma thromboxane, prostacyclin and endothelin in experimental hyperlipidemic rats

摘  要:用含2%月见草油的高脂饲料喂饲实验性高脂血症大鼠4周,分别对血脂、血清脂质过氧化物(LPO)、血浆超氧化物歧化酶(SOD)、血栓素(TXA2)、前列腺素(PGI2)及内皮素(ET)进行了检测,并用扫描电镜对动脉内皮进行了观察。结果发现:月见草油能降低血清总胆固醇(TC)及甘油三酯(TG)含量,提高高密度脂蛋白胆固醇(HDL-C)含量;月见草油能显著降低血清LPO水平,提高血浆SOD活力;月见草油组大鼠血浆PGI2水平明显增高,而TXA2/PGI2比值及血浆ET水平明显下降;月见草油组大鼠肝脏脂肪蓄积稍轻于高脂对照组;扫描电镜观察亦发现月见草油组大鼠动脉内皮损伤明显轻于高脂对照组。结果提示月见草油具有良好的降脂。The experiment was designed to elucidate the effect of Oenotheria binnis Seed Oil (OBSO) on experimental hyperlipidemia in rats. The contents of serum lipids, lipoperoxides (LPO), plasma superoxides dismutase (SOD), thromboxane (TXA 2), prostacyclin (PGI 2), endothelin (ET) and the ultrastructural pathological change of the aortic wall under the scanning electron microscope were observed. Hyperlipidemic Wistar rats were fed with high fat diets (HFD) containing OBSO (2%) for 4 weeks. The results showed that: OBSO could reduce the level of total serum cholesterol (TC) and triglyceride (TG), and increase the level of high density lipoprotein cholesterol (HDL C). OBSO could significantly reduce serum LPO level and markedly increase plasma SOD activities. The concentration of plasma PGI 2 in the group fed with OBSO was much higher than that in the group fed with HFD only, meanwhile the concentration of plasma ET and the ratio of TXA 2/PGI 2 in OBSO were lower than those in HFD group. Moreover, hepatic histopathological examinations showed that hepatic fat accumulation in OBSO group was less than that in HFD group. OBSO could attenuate the damage of aortic endothelium. These results indicate that OBSO is useful in treating hyperlipidemia, protecting endothelial cells and preventing atherosclerosis.
