IL-3对脐血体外扩增及裸鼠体内重建造血的影响

摘　　要：[目的]通过观察脐血单个核细胞(cord blood mononuclear cells,MNC)体外扩增及裸鼠体内重建造血情况,探讨IL-3在细胞因子组合中促进脐血扩增的作用。[方法]对同一份脐血采用两种细胞因子组合(TPO+FL+IL-6、TPO+FL+IL-6+IL-3)体外短期扩增9 d,采用流式细胞技术检测MNC及CD34+细胞数量变化。将扩增后MNC经尾静脉注入亚致死量照射的BABL/c裸鼠体内,观察裸鼠外周血象恢复情况,并采用多聚酶链式反应(PCR)及免疫组化技术检测植入指标。[结果]含IL-3的细胞因子组合对脐血MNC及CD34+细胞的扩增效果(8.96±0.52倍)优于不含IL-3的细胞因子组合(6.28±0.16倍)(P﹤0.01),扩增后的MNC在裸鼠体内可以重建造血并且形成植入,两组比较差异无显著性(P﹥0.05)。[结论]IL-3加入细胞因子组合可促进MNC体外增殖,且在裸鼠体内形成植入,重建造血。[Objective] To study the validation of interleukin-3 for use in the culture comprising different cytokines by observing the proliferative potential in vitro and the possibility to reconstitute the hematopoietic system in BABL/c mice of cord blood mononuclear cells. [Methods] After 9d suspension culturing of cord blood mononuclear ceils in two different cytokine combination （TPO＋FL＋IL-6, TPO＋FL＋IL-6＋IL-3 ）, the quantitative change of nucleated cells, CD34^＋ ceils were observed using flow cytometry. Using the techniques of polymerase chain reaction （PCR） and immunohistochemistry,, we examined the engrafting in the mice that had received tail-vein injection of the cord blood mononuclear cells. [ Results] The proliferative effect of cytokine combination including IL-3 （8.96 ± 0.52 floded） was higher than the other one （6.28 ± 0.16 ridded, P 〈 0.01 ）, It demonstrated significant difference （P 〈 0.05） . Their ability to reconstitute the hematopoietic system in BABL/c mice was similar and no significant difference in engraftment was found. [ Conclusion] Using IL-3 in cytokine combination could improve the proliferative potentials in vitro of cord blood mononuclear ceils and CD34^＋ ceils, and maintaining the ability to reconstitute the hematopoietic system in BABL/c mice.
