摘 要:目的探讨树突状细胞(DC)抗原负载方法。方法体外诱导培养外周血单核细胞来源的DC,分别用脂质体和Nucleo-fector转染试剂盒体外负载抗原,比较2种负载方法转染效率以及对DC表型的影响。结果Nucleofector转染方法能够显著的提高转染效率,2种转染方法对DC细胞的表型无显著影响。结论Nucleofector转染是一种有效的DC抗原体外负载方法,为抗原修饰的DC细胞疫苗的改进提供了新的途径。Objective To research the effective method of antigen transfection into dendritic cells (DC). Methods Human dendritic cells were derived from peripheral blood monocytes. The plasmid pEGFP-N1 was transfected into DC with liposome or by Nucleofeetor Kit. The transfeeting efficiency was detected by microscopy and FCM. Results The efficiency of Nucleofector Kit was significantly higher than that of liposome. No significant differences of the cellular morphology and surface molecules expression level of DC were shown between the transfections by liposome and Nucleofector Kit. Conclusion Nucleofector Kit is an effective method for antigen transfection and may provide a new strategy of tansfection for antigen-modified DC vaccine.
